Expression data in the absence of Notch1 in hair follicles

(Submitter supplied) Notch1 deficient hair matrix keratinocytes have lower mitotic rates, resulting in smaller follicles with fewer cells. In addition, the ratio of melanocytes to keratinocytes is greatly reduced. Microarray was performed to study downstream mechanism of Notch1-deficiency Keywords: genetic modification

Conditional activation of B-catenin in adult mouse epidermis for 0, 1, or 7 days

(Submitter supplied) Expression of deltaNB-cateninER allows the time and duration of B-catenin activation to be controlled precisely through the application of 4OHT. We have shown that B-catenin activation in adult mouse epidermis stimulates de novo hair follicles and the formation of a new bulge and melanocyte niche. We utilize Affymetrix Mouse Genome 430A 2.0 oligonucleotide arrays to investigate the differential regulation of RNA in skin from 7 week old female deltaNB-cateninER transgenic mice following 0, 1, or 7 days B-catenin activation in the epidermis. more...

Beta-catenin activation effect on skin: time course

Analysis of skin of deltaNB-cateninER transgenics following the activation of beta-catenin for up to 7 days. Onset and duration of beta-catenin activation in deltaNB-cateninER transgenics controlled by 4-hydroxytamoxifen. Results provide insight into how beta-catenin induces hair follicle growth.

Keratinocytes in embryos and newborns, and Notch1 expression in newborns

(Submitter supplied) Comparison between pattern of gene expression in uncultured keratinocytes derived from the epidermis of embryonic mice (E15.5) versus newborn mice and comparison between pattern of gene expression in primary keratinocytes derived from newborn mice plus/minus activated Notch1 expression. Keratinocyte differentiation promoted by Notch1 signaling

Comparison of gene expression in keratinocytes from embryos at 15.5 days of gestation to that from 2 day newborns infected with an adenovirus expressing a constitutively activate Notch1. Results indicate that Notch1 promotes differentiation in part through caspase 3.

Nipple fibroblast gene expression profile

(Submitter supplied) The nipple is an example of specialized epidermis. The unique epidermal stratification and gene expression is induced and maintained by developmental distinct fibroblast populations. To identify specific signaling modules involved in the maintenance of nipple epidermis we used gene expression profiling of purified fibroblast populations from a murine model of nipple skin formation

High proliferation and delamination during skin epidermal stratification

(Submitter supplied) The development of complex stratified epithelial barriers in mammals is initiated from single-layered epithelia. How stratification is initiated and fueled are still open questions. Previous studies on skin epidermal stratification suggested a central role for perpendicular/asymmetric cell division orientation of the basal keratinocyte progenitors. Here, we use centrosomes, that organize the mitotic spindle, to test whether cell division orientation and stratification are linked. more...

EGFR and hair shaft differentiation

(Submitter supplied) EGFR signaling controls skin development and homeostasis in mice and humans and its deficiency causes severe skin inflammation, which might affect epidermal stem cell behaviour. Here, we describe the inflammation-independent effects of EGFR-deficiency during skin morphogenesis and in adult HFSCs. Expression and alternative splicing analysis of RNAseq data from interfollicular epidermis and outer root sheath indicate that EGFR controls genes involved in epidermal differentiation, but also in centrosome function, DNA damage, cell cycle and apoptosis. more...

Global changes in gene expression in dermal fibroblasts with in vivo and in vitro deletion of the RBP-Jk gene

(Submitter supplied) It is currently unclear whether tissue changes surrounding multifocal epithelial tumors are a cause or consequence of cancer. Here, we provide evidence that loss of mesenchymal Notch/CSL signaling causes tissue alterations, including stromal atrophy and inflammation, which precede and are potent triggers for epithelial tumors. Mice carrying a mesenchymal-specific deletion of CSL/RBP-JK, a key Notch effector, exhibit spontaneous multifocal keratinocyte tumors that develop after dermal atrophy and inflammation. more...
