Circulating microRNA signature as novel biomarkers for osteoarthritis development

(Submitter supplied) Introduction: MicroRNAs (miRNAs) in circulation have emerged as promising biomarkers. In this study we aimed to identify a circulating miRNA signature for osteoarthritis (OA) patients. Methods: Serum samples were collected from 12 primary OA patients and 12 healthy individuals and were screened using the Agilent Human miRNA Microarray. Receiver Operating Characteristic (ROC) curves were constructed to evaluate the diagnostic performance of the deregulated miRNAs. more...

Microarray analysis of microRNA expression in cutaneous squamous cell carcinoma

(Submitter supplied) MicroRNAs (miRNAs) are a novel class of short RNAs which have shown to be dysregulated in a variety of cancers including squamous cell carcinoma (SCC) of the head&neck. Microarray based miRNA expression profiles of cutaneous SCC (cSCC) however have not been investigated so far. Seven patients with cutaneous SCC were enrolled in the study. Tumor biopsies (n=7) were taken from the center of the tumor. more...

Comparative microarray analysis of microRNA expression profiles in primary cutaneous malignant melanoma, cutaneous malignant melanoma metastases and benign melanocytic naevi

(Submitter supplied) Perturbations in microRNA (miRNA) expression profiles has been reported for cutaneous malignant melanoma (CMM). With regards to a rapidly growing number of newly discovered miRNA sequences, the availability of up-to-date miRNA expression profiles for primary cutaneous malignant melanoma (PCMM), cutaneous malignant melanoma metastases (CMMM) and benign melanocytic naevi (BMN) is limited. Patients with PCMM (n=9), CMMM (n=4) and BMN (n=8) were included in the study. more...

microRNA Expression in Normal and Diabetic Pancreatic Islets

(Submitter supplied) Genetic studies have identified ≥240 loci associated with type 2 diabetes (T2D), yet most of these loci lie in non-coding regions, masking the underlying molecular mechanisms. Recent studies investigating gene expression in pancreatic islets have provided key insights into the molecular drivers of T2D pathophysiology through comprehensive genetic and genomic analyses. However, similar studies investigating microRNA (miRNA) expression remain limited. more...

Mucosal gene expression profiling in (un)inflamed colon of patients with ulcerative colitis (UC)

(Submitter supplied) In this study, we investigated if miRNA expression in UC mucosa is altered and correlated our findings with mucosal mRNA expression. rs41291957 alters secondary RNA structure of the human pri-miR-143/145

(Submitter supplied) Purpose: To evaluate whether the single nucleotide polimorphsim rs41291957, located in the pri-miR-143/145, influences the primary RNA secondary structure. Methods: In vitro transcription and folding for the pri-miR-143/145 carrying the WT allele (G) or the mutated one (A) was carried out. The RNAs were then subjected to RNAseI treatement for 30 minutes and RNA seq performed on the digested nucleic acids. more...

Identification of 5'isomiR in HCC patients.

(Submitter supplied) The microRNA (miRNA) processing is a critical step for production of mature miRNAs. Its dysregulation leads to the increase of miRNA isoforms with heterogenous 5’ ends (isomiRs), recognizing distinct target sites with a shifted seed sequence. Although some miRNA genes display productive expression of their 5’-isomiRs in cancers, how the production is controlled and how they impact on tumor progression are unexplored yet. more...

Identification of novel serum biomarkers in early-stage HCC patients.

(Submitter supplied) Hepatocellular carcinoma (HCC) is one of the most common malignancies worldwide, and is the one of the few cancers in which a continued increase in incidence has been observed over several years. HCC associated with chronic liver disease evolves from precancerous lesion and early HCC to overt cancer, and identifying key molecules contributing to early stage HCC is an urgent need. α-Fetoprotein (AFP) is the best serum biomarker for diagnosis of HCC, but sensitivity is low, particularly in detection of early-stage HCC. more...

Effects of miR-4707-3p expression on differentiation of primary human neural progenitor cells

(Submitter supplied) To test the effects of miR-4707-3p expression on gene expression and differentiation of primary human neural progenitor cells (phNPCs), we produced two doxycycline inducible stable cell lines using lentiviral transduction. Lines expressed either mir-4707 and GFP or only GFP using the pTRIPZ expression vector. Stable lines were grown in differentiation media with doxycycline for one or two weeks before RNA was extracted and assayed on a Clariom S Pico Microarray Assay.

RIP: miR-202 mimic-transfected HeLa cells vs. negative control

(Submitter supplied) Potential miR-202 targets were identified using a targetome-wide RIP-based microarray. HeLa cells were transfected with either a miR-202 mimic or a scrambled single-stranded RNA negative control. RNA was isolated from total cell lysate prior to IP and from antibody-immobilized Protein G agarose beads-RNP complexes (post-IP).
