The effect of co-culture on MMTV-HER2 early lesion or primary tumor cell and alveolar macrophages [bulk RNA-seq]

(Submitter supplied) Disseminated cancer cells reside in the lung alveolar niche where they are exposed to interactions with alveolar macrophages. Here we report bulk RNA-sequencing data from co-cultured MMTV-HER2 mammary early lesion cells or mammary primary tumor cells cultured with lung alveolar macrophages. We discovered reciprocal regulation between mammary cells and alveolar macrophages, where early lesion cells instruct alveolar macrophages to maintain an anti-tumor phenotype and alveolar macrophages activate an epithelial to mesenchymal program in early lesion cells. more...

Extracellular vesicles derived from synovial inflammatory macrophages induce the onset of osteoarthritis in a FMRP-selectively sorted manner

(Submitter supplied) Osteoarthritis (OA) is an aging-related degenerative joint disease without effective therapeutic. In the early stage of OA, mild synovitis has been proven to induce normal cartilage lesions. However, the mechanism is poorly defined. Here, we identified that the extracellular vesicles (EVs) derived from synovial inflammatory macrophages regulate the autophagy function of chondrocytes, acting as the dominant inducer of the onset of cartilage degeneration in normal and OA joints. more...

Acute IL-6 exposure triggers canonical IL6Ra signaling in hiPSC microglia, but not neural progenitor cells

(Submitter supplied) Background: Prenatal exposure to elevated interleukin (IL)-6 levels is associated with increased risk for psychiatric disorders with a putative neurodevelopmental origin, such as schizophrenia (SZ), autism spectrum condition (ASC) and bipolar disorder (BD). Although rodent models provide causal evidence for this association, we lack a detailed understanding of the cellular and molecular mechanisms in human model systems. more...

IRF4 contributes to chemoresistance in IGH-BCL2 positive diffuse large B-cell lymphomas by mediating BCL2 induced SOX9 expression

(Submitter supplied) SOX9 was identified as a prognostic biomarker particularly in IGH-BCL2 translocated germinal center B cell (GCB) subtypes of the diffuse large B cell lymphoma (DLBCL) and plays a vital role in lymphomagenesis. However, the molecular mechanism that modulating the aberrant expression of SOX9 in this DLBCL subset remains unknown. We have demonstrated that SOX9 enhanced IGH-BCL2 positive DLBCL subset resistance to either chemotherapy or BCL2 inhibitor. more...

Piezo1-mediated Mechanotransduction Shapes ILC2 Translational Activity, Functions, and Lung Pathogenicity

(Submitter supplied) Group-2 innate-lymphoid cells (ILC2s) are critical mediators of the type-2 immune responses in multiple lung pathologies. We show that Piezo1, a mechanosensitive ion channel, plays a key role in regulating ILC2 functions by linking mechanical cues to biochemical signaling pathways. Both murine and human ILC2s strongly express Piezo1, and its activation by Yoda1 selectively enhances IL-13 production through calcium influx, which activates the mTOR-S6K pathway. more...

PARP-1 as a potential therapeutic target in ERα-positive endocrine therapy-resistant breast cancer

(Submitter supplied) Abstract Several endocrine therapy (ET) resistance mechanisms for ER-positive (ER+) breast cancer (BC) have been proposed, including acquired ESR1 (ERα gene) mutations. The two most common ESR1 mutations are Y537S and D538G, which give rise to a constitutively active receptor with reduced affinity for agonists and antagonists. The discovery of new effective therapies remains a significant challenge in treating mutated ER+ BC. more...

Effects of Pd1 targeted myeloid deletion on brain development

(Submitter supplied) During early brain development, the nervous system continues to evolve and improve, and cells in different stages contact and connect to form a unique nervous system network. Various cross-talk between cell populations is the key to neurological homeostasis. Communication between astrocytes and myeloid cells affects brain function. Programmed cell death protein 1 (PDCD1), also known as PD-1 or CD279 (differentiation cluster 279), is a member of the immunoglobulin gene superfamily. more...

Combining multiple stressors unexpectedly blocks bacterial migration and growth

(Submitter supplied) In nature, organisms experience combinations of stressors. However, laboratory studies use batch cultures, which simplify reality and focus on population-level responses to individual stressors. In recent years, bacterial stress responses have been examined with single-cell resolution using microfluidics. Here, we use a microfluidic approach to simultaneously provide a physical stressor (shear flow) and a chemical stressor (H2O2) to the human pathogen Pseudomonas aeruginosa. more...

Effets of plant compounds on estrogen receptor positive breast cancer cell line MCF-7

(Submitter supplied) This study was designed to determine the effects of plant derived compounds on the transcriptome of estrogen receptor positive breast cancer. Phytoestrogens are compounds produced by plants that possess weak estrogenic activity. They can be found in a wide variety of plants, including fruits and vegetables, but are most abundant in leguminous plants. Legumes are consumed in almost every diet throughout the world, and in addition to the seeds, many other parts of the plant are also edible, including the pods of some varieties. more...

Aspartate signaling increases the aggressiveness of lung metastases by inducing eIF5A-mediated translation (Total RNA-Seq)

(Submitter supplied) Lung metastases are detected in more than half of patients with metastatic tumors. However, it remains largely unknown why the lung environment is a permissive niche for metastases. Here, we discover that pulmonary aspartate triggers a cellular signaling cascade in disseminated cancer cells resulting in a translational program that boosts lung metastasis. Specifically, we observe that patients and mice with breast cancer have high concentrations of aspartate in their lung interstitial fluid. more...

3D-Printed PCL Scaffolds Loaded with bFGF and BMSCs(PCLMF) Enhance Tendon-Bone Healing in Rat Rotator Cuff Tears by Immunomodulation and Osteogenesis Promotion

(Submitter supplied) Rotator cuff tears are the most common conditions in sports medicine and attract increasing attention. Scar tissue healing at the tendon-bone interface results in a high rate of retears, making it a major challenge to enhance the healing of the rotator cuff tendon-bone interface. Biomaterials currently employed for tendon-bone healing in rotator cuff tears still exhibit limited efficacy. 3D printing, a promising technology, enables the customization of scaffold shapes and properties. more...
