Fetal and neonatal ductus arteriosus and aorta: endothelial cells

Analysis of endothelial cells (ECs) isolated from the ductus arteriosus (DA) and aorta of fetuses and neonates. The DA, a fetal shunt artery between the pulmonary artery and the aorta, closes immediately after birth. Results provide insight into EC involvement in DA-specific morphology and function.

Expression data from endthelial cells in ductus arteriosus and aorta in rat fetuses or neonates

(Submitter supplied) Endothelial cells (Ecs) lining the blood vessels have been known to have a variety of functions and play a central role in homeostasis of the circulatory system. Since ductus arteriosus (DA) has unique characteristics comparing other vessels, we hypothesized that ECs were the primary target to investigate the molecular mechanisms of DA-specific vasoconstriction and vascular remodeling.

Dlx1 and Rgs5 in the Ductus Arteriosus: Vessel-specific Genes Identified by Transcriptional Profiling of Laser-capture Microdissected Endothelial and Smooth Muscle Cells

(Submitter supplied) Closure or patency of the ductus arteriosus is a critical event in neonatal life. We aimed to identify genes that are specifically expressed in the ductus arteriosus versus (the non-closing) aorta Gene expression profiling of laser-captured microdissected cells offers the opportunity to study gene expression profiles in cells of different embryonic origin

Transcriptional profiles in the chicken ductus arteriosus during hatching

(Submitter supplied) The ductus arteriosus constricts after birth or hatching and eventually closes to terminate embryonic circulation. Chicken embryos have two long ductus arteriosus. Then the pulmonary artery-sided and descending aorta-sided ductus arteriosus have distinct functional characteristics, such as oxygen responsiveness. We used microarrays to elucidate the difference between the pulmonary artery-sided and descending aorta-sided ductus arteriosus in their transcriptional profiles.

Expression profiling in perinatal Ductus Arteriosus (DA) and Aorta (Ao).

(Submitter supplied) Compared the gene expression profiles of two contiguous vessels pre and post - birth, namely the Ductus Arteriosus (DA), committed to become a fibrous ligament, and the Aorta (Ao), which conversely takes on a permanent activity. The effect of artificially enhanced O2 tension was analyzed in DA in the pre-birth condition. Keywords: Comparison gene expression

Gene Profiling in Mouse Fetal Ductus Arteriosus

(Submitter supplied) The ductus arteriosus (DA) is a fetal vascular shunt that is located between the main pulmonary artery and the aorta. Oxygenated fetal blood from the placenta is shunted past the uninflated fetal lungs, crosses the DA, and is then available to the peripheral organs. In utero closure of the DA is deleterious, but postnatal closure of the DA is necessary for establishment of pulmonary circulation and the transition to newborn life. more...

Microarray gene expression analysis in ovine ductus arteriosus during fetal development and birth transition.

(Submitter supplied) Patent ductus arteriosus (PDA) in the newborn is the most common congenital heart anomaly, and is significantly more common in preterm infants. Contemporary pharmacological treatment is effective in only 70 to 80% of the cases. Moreover, indomethacin or ibuprofen, which are used to close a PDA may be accompanied by serious side effects in premature infants. To explore the novel molecular pathways, which may be involved in the maturation and closure of the ductus arteriosus (DA), we used fetal and neonatal sheep to test the hypothesis that maturational development of DA is associated with significant alterations in specific mRNA expression.

Human Cavernosal Endothelial Cell Phenotype

(Submitter supplied) Purpose: To identify the molecular phenotype of endothelial cells (EC) isolated from the unique vasculature of the corpus cavernosum. Methods: Human EC derived from corpus cavernosum (HCCEC, n=5), coronary artery (HCAEC, n=4) and umbilical vein (HUVEC, n=3) were grown in culture and mRNA transcripts quantified by Affymetrix GeneChip microarrays. Genes differentially expressed across samples were partitioned around medoids to identify genes with highest expression in HCCEC. more...

Endothelial cell diversity revealed by global expression profiling

(Submitter supplied) The vascular system is locally specialized to accommodate widely varying blood flow and pressure and the distinct needs of individual tissues. The endothelial cells (ECs) that line the lumens of blood and lymphatic vessels play an integral role in the regional specialization of vascular structure and physiology. However, our understanding of EC diversity is limited. To explore EC specialization on a global scale, we used DNA microarrays to determine the expression profile of 53 cultured ECs. more...

Endothelial cell-enriched genes expression in mouse embryo

(Submitter supplied) The early blood vessels of the embryo and yolk sac in mammals develop by aggregation of de novo forming angioblasts into a primitive vascular plexus, which then undergoes a complex remodeling process. Angiogenesis is also important for disease progression in the adult. However, the precise molecular mechanism of vascular development remains unclear. It is therefore of great interest to determine which genes are specifically expressed in developing endothelial cells. Here, we utilized Flk1-deficient mouse embryos, which lack endothelial cells, to perform a genome-wide survey for genes related to vascular development.

In vivo endothelial transcriptional profiles of porcine coronary and iliac arteries

(Submitter supplied) Phenotypic heterogeneity among arterial ECs is particularly relevant to atherosclerosis since the disease occurs predominantly in major arteries, which vary in their atherosusceptibility. To explore EC heterogeneity, we used DNA microarrays to compare gene expression profiles of freshly harvested porcine coronary and iliac artery ECs. We demonstrate that in vivo the endothelial transcriptional profile of a coronary artery (the right coronary artery) is intrinsically different from that of a major conduit vessel (the external iliac artery), and that this difference is consistent with former vessel being more prone to atherosclerosis. more...
