Comparative transcriptional analysis identifies genes associated with the attenuation of Theileria parva infected cells after long-term in vitro culture

(Submitter supplied) Administration of attenuated autologous Theileria parva infected cells can be used as an alternative to the infection-and-treatment method for inducing immunological protection against East Coast Fever. The mechanism of attenuation however has not been described. Using RNA sequencing, the transcriptomes of both host and parasite in uninfected (control), pathogenic (day 7 post-infection) and attenuated (day 69 post-infection) T. more...

Comparison of high throughput RNA sequences between Babesia bigemina and Babesia bovis revealed consistent differential gene expression that is required for Babesia life cycle in the vertebrate and invertebrate hosts.

(Submitter supplied) To understand Babesia gene regulation during tick and mammalian host infection, we performed high throughput RNA-sequencing using samples collected from calves and Rhipicephalus microplus ticks infected with Babesia bigemina. We evaluated gene expression differences between B. bigemina kinetes and blood-stage parasites

Differential transcriptional expression by Babesia bovis during parasite development within bovine erythrocytes and tick hemolymph

(Submitter supplied) Babesia parasites transition between a mammalian host, where they cause babesiosis, and the tick vector that transmits them. This transition provides an environmental signal resulting in altered gene expression allowing the completion of the parasite’s life cycle. A comparison of the different life stages that occur within mammalian and tick hosts can provide insight into the adaptation of Babesia to these different environments. more...

ApiAP2 Factors as Candidate Regulators of Stochastic Commitment to Merozoite Production in Theileria annulata

(Submitter supplied) Investigation of parasite (T. annulata) gene expression over the course of the life-cycle (sporozoite->macroschizont->merozoite->piroplasm). The study focused on the expression of known and putative transcription factors, in particular members of the ApiAP2 gene family. Up-stream motifs associated with stage-specifically expressed genes were identified during the course of the analysis.

Morphological and molecular descriptors of the developmental cycle of Babesia divergens parasites in human erythrocytes

(Submitter supplied) Human babesiosis, especially caused by the cattle derived Babesia divergens parasite, is on the increase, resulting in renewed attentiveness to this potentially life threatening emerging zoonotic disease. The molecular mechanisms underlying the pathophysiology and intra-erythrocytic development of these parasites are poorly understood. This impedes concerted efforts aimed at the discovery of novel anti-babesiacidal agents. more...

A bovine lymphosarcoma cell line infected with Theileria annulata exhibits an irreversible reconfiguration of host cell gene expression

(Submitter supplied) The experiment investigates bovine gene expression in response to BW720c treatment in uninfected and Theileria annulata-infected cell cultures Theileria annulata, an intracellular parasite of bovine lymphoid cells, induces substantial phenotypic alterations to its host cell including continuous proliferation, cytoskeletal changes and resistance to apoptosis. While parasite induced modulation of host cell signal transduction pathways and NFκB activation are established, there remains considerable speculation on the complexities of the parasite directed control mechanisms that govern these radical changes to the host cell. more...

Expression analysis of the Theileria parva subtelomere-encoded variable secreted protein gene family

(Submitter supplied) The protozoan parasite Theileria parva infects and transforms bovine lymphocytes inducing uncontrolled proliferation. The transforming schizont resides free in the host cell cytoplasm and it is assumed that proteins released from the parasite contribute to host cell transformation and parasite persistence. The identification and characterisation of parasite genes encoding candidate secreted proteins constitutes a first step towards elucidating this complex process. more...

Agilent-036891 Babesia bovis_UP array

(Submitter supplied) Custom B. bovis microarray slides were manufactured using a proprietary non-contact industrial inkjet printing process, in which oligo monomers were deposited uniformly onto specially-prepared glass slides. This in situ synthesis process printed 60-mer length oligonucleotide probes, base-by-base, from digital sequence files. The precise inkjet process enabled the delivery of extremely small, accurate volumes (picoliters) of the chemicals to be spotted. more...

Custom Theileria parva oligonucleotide microarray

(Submitter supplied) Microarray design and printing was performed by Dr. Simon Kang’a at The Institute for Genomic Research (TIGR), now the J. Craig Venter Institute (JCVI). A total of 4060 unique 70 mer oligonucleotides were designed from an initial 4082 predicted ORF of the Theileria parva (Muguga) genome using the Pick70 program of ArrayOligoSelector software (http://arrayoligosel.sourceforge.net). Oligonucleotide design considerations included uniqueness, avoidance of internal self-annealing structures, narrow annealing temperature (Tm) range over the entire oligonucleotide set, masking of low-complexity regions, balanced base composition (28-30%GC) and proximity to the 3' end. more...
