Silencing of GATA3 defines a novel stem cell like subgroup of ETP-ALL

(Submitter supplied) Genome wide expression profiling was used to detect cases of adult T-ALL lacking GATA3 expression. GATA3low T-ALL exhibited enrichment of myeloid/lymphoid progenitor (MLP) and granulocyte/monocyte progenitor (GMP) genes, while T-cell specific signatures were downregulated. Among upregulated genes FLT3 was identified and mutational analyses revealed a high rate of FLT3 mutations.

Loss of the FAT1 tumor suppressor promotes resistance to CDK4/6 inhibitors via the Hippo pathway

Probing the Canonicity of the Wnt/Wingless Signaling Pathway [STARR-seq]

(Submitter supplied) The hallmark of canonical Wnt signaling is the transcriptional induction of Wnt target genes by the beta-catenin/TCF complex. Several studies have proposed alternative interaction partners for beta-catenin or TCF, but the relevance of potential bifurcations in the distal Wnt pathway remains unclear. Here we study on a genome-wide scale the requirement for Armadillo (Arm, homolog of beta-catenin) and Pangolin (Pan, Drosophila’s TCF) in the Wnt/Wingless (Wg)-induced transcriptional response of Drosophila Kc cells. more...

COLO320 DNA copy number analysis

(Submitter supplied) Somatic DNA alteration underlies tumor development and progression, and gives rise to tumors with diverse genetic contexts. Here, we identify in a collection of 29 colorectal cancer cell lines and 226 primary colorectal tumors recurrent amplification of chromosome 13, an alteration highly restricted to colorectal-derived cancers. A minimal region of amplification on 13q12.2 pinpoints caudal type homeobox transcription factor CDX2, a master regulator of anterior-posterior patterning, midgut development, and intestinal epithelial cell differentiation and maintenance. more...

A common set of DNA regulatory elements shapes Drosophila appendages

(Submitter supplied) We mapped open chromatin by FAIRE-seq and measured gene expression by RNA-seq in 3 types of Drosophila samples: staged whole embryos, imaginal discs, pharate appendages. We first demonstrate that regions of open chromatin precisely define regions of enhancer activity in developing embryos. In contrast to the dynamic changes in open chromatin observed between different stages of embryogenesis, we found that the open chromatin profiles in wing, leg, and haltere imaginal discs are nearly identical. more...
