Characterized functions of hypothetical genes identified by Tn-seq important for pathgoenesis of Staphylococcus aureus pneumonia

(Submitter supplied) To identify new genetic elements required for S.aureus pathogenesis, we performed a transposon sequencing in the context of acute murine penumonia. We screened conditionally essential genes identified by Tn-seq and confirmed 6 previously uncharacterized genes were important during infection. As further study, we performed RNA-seq to identify uncharacterized genes.

Transcriptome Comparison of phoU Homologies: Genes Deletion strain and the Parent Strain in stationary phase (10 h)

(Submitter supplied) Purpose: To compare the transcriptome changes in phoU1(serp0956) or phoU2(serp0316)deletion strain with the parent strain SE1457 in stationary phase (10 h). Methods: Total RNA was isolated and sequenced from the phoU1 deletion strain, phoU2 deletion strain and the parent strain SE1457 in stationary phase (10 h) in triplicate using an illumina high-seq 2500. Raw data was analyzed using TopHat. Genes were considered changed which performed fold-change>=1.5 and FDR<=0.05. more...

Interplay of CodY and CcpA in regulating central metabolism and biofilm formation in Staphylococcus aureus

Identification of a novel LysR-type transcriptional regulator in Staphylococcus aureus that is crucial for secondary tissue colonization during metastatic bloodstream infection

(Submitter supplied) Purpose: The goal of this study was to identify Staphylococcus aureus genes which are important during establishment of metastatic infections in bloodstream infections. Methods: Pooled mariner transposon mutant libraries were generated as previously described (PMID: 27185949). The libraries were sequenced on the Illumina HiSeq 2500 platform with the transposon-specific oligonucleotide primer Himar1-Seq. more...

Staphylococcus aureus walKR mutants (TPS3130 and TPS3190) vs. parent strains (JKD6009 and JKD6004)

(Submitter supplied) WalKR is an essential two component regulatory system in S. aureus, thought to control cell wall metabolism. Using genome sequencing of 5 paired clinical isolates of vancomycin-susceptible and vancomycin-intermediate S. aureus we found frequent, but unique, mutations in this locus. To investigate the contribution of these mutations to vancomycin resistance allelic replacement WalK (G223D) and WalR (K208R) mutants were generated and compared to the parent strains. more...

Staphylococcus aureus SH1000 compared to SH1000 ΔthyA

(Submitter supplied) Staphylococcus aureus thymidine-dependent small-colony variants (TD-SCVs) are frequently isolated from patients with chronic S. aureus infections after long-term treatment with trimethoprim-sulfamethoxazole (TMP-SMX). In TD-SCVs, mutations of thymidylate synthase (thyA, TS), essential for DNA synthesis, occur. However, it has never been shown, that TMP-SMX is responsible for the induction and selection of TD-SCVs. more...
