MicroRNA profiling of human pancreas development.

(Submitter supplied) MicroRNAs (miRNAs) are non-coding RNAs that play a fundamental role in regulation of gene expression affecting differentiation and development. In particular, miRNAs have been described to regulate genes important for pancreatic development and islet function. The aim of this study was to determine the miRNA expression signature during human pancreas development. We identified 212 miRNAs that were expressed throughout different gestational ages. more...

Expression data from human embryonic (9-12w) and post-natal livers

(Submitter supplied) The liver is a multifunctional organ, which undergoes rapid changes during the developmental period and relies on tightly-regulated gene expression. Little is known regarding the complex expression patterns of mRNAs during the early stages of human liver development in comparison to post-natal livers. We used microarrays to detail the global programme of gene expression during liver development following specification, at the stage of proliferation and differentiation when compared to post-natal livers, and identified distinct classes of up-regulated and downregulated genes during this process.

The human reticulocyte transcriptome (HG-U133_Plus2.0)

(Submitter supplied) RNA from circulating blood reticulocytes was utilized to provide a robust description of genes transcribed at the final stages of erythroblast maturation. After depletion of leukocytes and platelets, Affymetrix HG-U133Plus 2.0 arrays were hybridized with probe from total RNA isolated from blood sampled from 6 umbilical cords and 6 healthy adult humans.

Let-7 microRNAs are developmentally regulated in circulating human erythroid cells

(Submitter supplied) MicroRNA are ~22nt-long small non-coding RNA that negatively regulate protein expression through mRNA degradation or translational repression in eukaryotic cells. Based upon their importance in regulating development and terminal differentiation in model systems, erythrocyte microRNA profiles were examined at birth and in adults to determine if changes in their abundance coincide with the developmental phenomenon of hemoglobin switching. more...

Fetal and adult reticulocytes

Comparison of reticulocytes from adult peripheral blood and umbilical cord blood. Hemoglobin (Hb) composition switches around the time of birth from fetal Hb to adult Hb. Together with microRNA profiling, results provide insight into the role of microRNAs in fetal-to-adult Hb switching.

Genomic profiling of microRNA and messenger RNA reveals deregulated microRNA expression in prostate cancer.

(Submitter supplied) MicroRNAs are small non-coding RNAs that regulate mRNA function. Recent studies have shown that microRNA expression is altered in tumors. We studied the expression of both microRNAs and mRNAs in 60 primary prostate tumors and 16 non-tumor prostate tissues to evaluate the involvement of microRNAs in prostate cancer. Global microRNA expression was determined in RNA isolated from fresh-frozen human tissues with a custom oligonucleotide microarray chip. more...

RIP-Chip analysis supports different roles for AGO2 and GW182 proteins in recruiting and processing microRNA targets (microRNA)

(Submitter supplied) MicroRNAs are small non-coding RNA molecules mediating the translational repression and degradation of target mRNAs in the cell. Mature miRNAs are used as a template by the RNA-induced silencing complex (RISC) to recognize the complementary mRNAs to be regulated. To discern further RISC complex functions, we analyzed the activities of two RISC proteins, AGO2 and GW182, in the MCF-7 human breast cancer cell line. more...

Profiling and Functional Analyses of MicroRNAs and Their Target Gene Products in Human Uterine Leiomyomas

(Submitter supplied) Human uterine leiomyomas (ULM) are characterized by dysregulation of a large number of genes and non-coding regulatory microRNAs. In order to identify microRNA::mRNA associations relevant to ULM pathogenesis, we examined global correlation patterns between the altered microRNA expression and the predicted target genes in leiomyomas and matched myometria. We found that some but not all of the predicted and inversely correlated target genes in ULMs can be directly regulated by microRNAs in vitro. more...

Profiling the transcriptome: synaptoneurosomes capture the molecular effects of alcohol consumption

(Submitter supplied) Action of alcohol on synaptic mRNA in the amygdala of mice Chronic alcohol consumption induces changes in gene expression, causing persistent long-term neuro-adaptations and the remodeling of synaptic structures. These alcohol-induced synaptic changes may rely specifically on the local translation of mRNAs in the synaptic compartments of the cell. We profiled the transcriptome from synaptoneurosomes (SN) and paired total homogenates (TH) of amygdala to analyze the synaptic adaptations induced by chronic voluntary alcohol consumption in mice. more...

Regulation of the human placental (pro)renin receptor-prorenin-angiotensin system by microRNAs

(Submitter supplied) The placental renin-angiotensin system (RAS) is important for placentation. RAS expression is greatest in early gestation. This may be due (in part) to suppression of miRNAs that target the placental RAS, but this has never been explored. In this study, human placental miRNAs were measured at 10–11 (early), 14–18 (mid), and 38–40 (term) weeks gestation, as well as in placentae from women with early- or late-onset preeclampsia (n=4/group), using an Agilent miRNA microarray (V19). more...

Expression data from human embryonic stem cell differentiation

(Submitter supplied) MicroRNAs (miRNAs) are noncoding RNAs of approximately 22 nucleotides in length that usually suppress the translation of target messenger RNAs (mRNAs) through partial complementarity to the 3¡¦ untranslated region (3¡¦ UTR) of protein-coding mRNAs in animals. However, there is increasing evidence that miRNAs can also reduce the steady-state levels of their target mRNAs in animals. In this investigation, both miRNA and mRNA profiles from the undifferentiated human embryonic stem cell line hES-T3 (T3ES), hES-T3 derived embryoid bodies (T3EB) and hES-T3 differentiated fibroblast-like cells (T3DF) were quantitatively determined. more...

Expression data from primary term human decidual cells treated with interleukin-1-beta for 6 hours.

(Submitter supplied) Preterm birth is an important unsolved clinical problem. Despite advanced treatments, infants who survive prematurity remain at increased risk for permanent disabilities. In approximately one-third of cases, prematurity is related to infection and/or inflammation, which renders hostile the normally receptive intrauterine environment. Proinflammatory cytokines provoke up-regulation of genes that promote uterine contractions. more...
