Influenza virus infection-induced gene expression changes of regional B cells are mediated in part through type I IFN

(Submitter supplied) Influenza virus infection-induced gene expression changes of regional B cells are mediated at least in part through type I Interferon: Our objective is to determine whether the influenza virus-infection induced gene expression changes in regional lymph node B cells are facilitated at least in part through type I interferon. Our specific aim is to compare the gene expression profile of highly FACS-purified B cells in the regional lymph nodes of wildtype and IFNR-/- mice prior to and 48h following infection with influenza virus infection and to contrast this expression profile with that of FACS-purified wildtype B cells activated in vitro with IFN-beta +/- anti-CD86 for 12h. more...

Interferon receptor deficient lymph node B cell response to influenza infection

Analysis of lymph node B cells from interferon receptor (IFNR) null mutants infected with the influenza virus. Wild type B cells treated with IFN-beta also examined. Results provide insight into the role of IFNR signaling in the immune response mediated by B cells during influenza infections.

Gene profiling analysis of mouse splenic DC subpopulations

(Submitter supplied) To characterize differences between BALB/c splenic CD11cintB220+Gr1+ PDCs (plasmacytoid dendritic cells), CD11cintB220+CD49b+ IKDCs (interferon producing killer-dendritic cells), and CD11chighB220- cDCs (conventional dendritic cells), we performed gene expression profile analysis using Affymetrix chips. We FACS-sorted BALB/c spleen DC subpopulations. Comparison of differentially expressed genes between IKDCs and cDCs vividly revealed selective expression of multiple NK-related genes in IKDCs . more...

Dendritic cell subpopulations: spleen (MG-U74C)

Analysis of three dendritic cell (DC) subpopulations from BALB/c spleens: interferon-producing killer DCs (IKDCs), plasmacytoid DCs (PDCs), and conventional DCs (cDCs). Results identify IKDCs as a distinct DC lineage and provide insight into the role of IKDCs in linking innate and adaptive immunity.

Transcription profile of B cells isolated from peripheral lymph nodes of wild-type vs CCR7-/- mice

(Submitter supplied) We investigated the transcriptional profile of B cells isolated from peripheral lymph nodes of CCR7-/-C57BL/6 and C57BL/6 mice respectively. B cells were sorted as follows: CD19+, CD3-, CD11c-, CD11b-, NK1.1-, GR-1-. Very few genes were differentially regulated in CCR7-/- B cells, however the expression of a number of genes associated with B-cell activation was increased in CCR7-/- B cells compared to WT B cells. more...

Single-cell RNA-seq for identification of B-cell specific checkpoint molecules regulating anti-tumor immunity

(Submitter supplied) The role of B cells in anti-tumor immunity is still debated and accordingly, most therapies have focused on targeting T and NK cells to inhibit tumor growth1,2. Here, using high-throughput flow cytometry, bulk and single-cell RNA- and BCR-sequencing of B cells temporally during B16F10 melanoma growth, we identified a subset of B cells that expands specifically in the draining lymph node over time in tumor bearing-mice. more...
