Targeted DamID Detects Cell Type Specific Histone Modifications in intact tissues or organisms

(Submitter supplied) Histone modifications play a key role in regulating gene expression and cell fate during development and disease. Current methods for cell-type specific genome-wide profiling of histone modifications require dissociation and isolation of cells and are not compatible with all tissue types. Here we adapt Targeted DamID to recognise specific histone marks, by fusing chromatin binding proteins or single-chain antibodies to Dam, an E. more...

Identification of genes regulated by mechanical tension in Xenopus laevis explants by transcriptome comparison

(Submitter supplied) During gastrulation and neurulation, the chordamesoderm and overlying neuroectoderm of vertebrate embryos converge under the control of a specific genetic programme to the dorsal midline, simultaneously extending along it. However, whether mechanical tensions resulting from these morphogenetic movements play a role in long-range feedback signalling that in turn regulates gene expression in the chordamesoderm and neuroectoderm is unclear. more...

H3K4 Methylation-Mediated Memory of an Active Transcriptional State Impairs Nuclear Reprogramming

(Submitter supplied) Xenopus eggs can induce the reversal of differentiation processes of somatic cells. Yet, the egg is not fully efficient in reprogramming a differentiated nucleus, as certain genes retain a memory of gene expression of their somatic cell of origin. This is thought to be a reason for the low success rate of current cloning and reprogramming strategies. While previous studies addressed extensively the mechanisms that maintain an inactive state of genes (OFF-memory), we investigated the importance of memory of an active transcriptional state (ON-memory) in maintaining cell fate identity and on resistance to reprogramming. more...

Methylome analysis of deoxyadenosines in higher eukaryotes

(Submitter supplied) Here, we report that we detected N-6-methyl-deoxyadenosine (dA6m) not only in frog DNA, but also in other species including mouse and humans. Our methylome analysis revealed that dA6m is widely distributed across the eukaryotic genome, and it is present in different cell types. dA6m is commonly depleted in gene exons, whilst it remains abundant in introns. Finally, we have identified the putative consensus sequence, where there is preferential methylation. more...
