Effect of Gram Negative Anaerobic Bacteria on Normal Human Skin Keratinocytes (HEKa cells)

(Submitter supplied) Objectives: To define the inflammatory signature of healthy keratinocytes induced by gram-negative anaerobe bacteria commonly found in HS and investigate pathways of activation. Methods: Type strains of P. nigrescens, P. melanogenica, P. intermedia, P. asaccharolytica, F. nucleatum, as well as S. aureus and the normal skin commensal S. epidermidis were heat-killed and co-incubated with normal human keratinocytes. more...

Dysregulation of mixed meal-induced gene expression in skeletal muscle in obesity and type 2 diabetes

(Submitter supplied) Physical inactivity and western diet are the main factors leading to skeletal muscle (and whole-body) insulin resistance – a defect in insulin-mediated glucose uptake and metabolism – and associated metabolic diseases, such as obesity and type 2 diabetes mellitus (T2D). The study of the response to a mixed meal (normalized for body mass and/or basal metabolic rate) is of particular interest, as it simulates the physiological response to a typical (daily) single meal that depends on both the secretory function of the beta cells (insulin response) and the sensitivity of skeletal muscle signaling to insulin – variables, which markedly changing during development of insulin resistance and T2D. more...

High-density genetic map and genome-wide associaation studies of floral color traits in Phalaenopsis orchids

(Submitter supplied) The F1 population of Phalaenopsis Intermedia, which established from the cross between P. equestris and P. aphrodite was applied to build a high-density genetic map based on SSRs and SNPs from GBS methodology, with the digestion of by Hinp1 I and Hae III. In addition, another GBS was performed with enzymes ApeK I and Hae II to increase the SNP number for the GWAS analysis. We identified 10 SNPs highly associated with floral aesthetic trait, and among them, 4 were associated with flower color related. more...

Changes in gut microbiota induced by oral pathobiont deteriorate the pathology of non-alcoholic fatty liver disease

(Submitter supplied) Periodontitis increases the risk of non-alcoholic fatty liver disease (NAFLD). However, the precise mechanisms are unclear. Here, gut dysbiosis induced by orally administered Porphyromonas gingivalis, a representative periodontopathic bacterium, was implicated in the deterioration of NAFLD pathology. C57BL/6 mice were administered with the vehicle, P. gingivalis or Prevotella intermedia, with weaker periodontal pathogenicity, followed by feeding on a choline-deficient, high fat diet (CDAHFD60). more...

Gender-specific transcriptional profiles identified in β-thalassemia patients

(Submitter supplied) Lifelong treatment and major complications characterize β-thalassemia patients, whether they suffer from the milder type of the disease (β-thalassemia intermedia, TI) or the severe type (β-thalassemia major, TM). This study aimed to ascertain the differences in the transcriptional profiles of TI and TM patients to identify targets for stratification and the development of personalized therapeutic strategies. more...

Gene Expression In Blood From an Individual With β-Thalassemia: an RNA Sequence Analysis

(Submitter supplied) Background: The thalassemias are highly diverse at both the molecular and clinical levels. Many of the HBB mutations that result in β-thalassemia are missense mutations in the coding region of the β-globin gene, but a few cause alternative splicing, and interfere with normal processing of the β-globin transcripts. Transcriptome profiling in individuals affected with β-thalassemia, especially in individuals who carry novel mutations in the HBB, may improve our understanding of the heterogeneity and molecular mechanisms of the disease. more...

Method for absolute quantification of microbial communities by using both microarrays and competitive PCR

(Submitter supplied) We investigated an improved method that combines competitive PCR and microarray techniques. This approach allowed us to quantify specific bacterial groups mounted on DNA chips with accuracy close to that of real-time PCR, despite a measurement at the end point of PCR, and also to estimate the bacterial DNA content in sample DNA.

Peripheral Blood Transcriptomic Analysis for Salmonella Infection in Pig

(Submitter supplied) Peripheral blood transcriptome is an important intermedia data source for investigation of the mechanism of Salmonella invasion, proliferation, and transmission but its development in pig is quite limited. We challenged four weeks old piglets (Duroc × Landrace × Yorkshire crossbred) with Salmonella enterica serovar Typhimurium LT2 and investigated the peripheral blood gene expression profile before treatment (d0) and at 2 and 7 day post inoculation (dpi) using deep sequencing technology.

Identification of microRNAs in Caragana intermedia by high-throughput sequencing and expression analysis of 12 microRNAs and their targets under salt stress

(Submitter supplied) MicroRNAs (miRNAs) are endogenous small RNAs that play important roles in various biological and metabolic processes in plants. Caragana intermedia is an important ecological and economic tree species that is prominent in the desert environment of west and northwest China. To date, no investigation into C. intermedia miRNAs has been reported. In this study, high-throughput sequencing of small RNAs was performed to identify both conserved and novel miRNAs, and also their target mRNA genes in C. more...

Transcriptional profile in low to high-level macrolide resistant C. jejuni strain selected from NCTC 11168

(Submitter supplied) Campylobacter, a major foodborne pathogen, is increasingly resistant to macrolide antibibotics. Previous findings suggeted that development of macrolide resistance in campylobacter requires a multi-step process, but the molecular mechanisms involved in the process are not known. In our study, multiple series of macrolide-resistant C. jejuni mutants were selected in vitro by stepwise exposure of C. jejuni NCTC11168 to increasing concentrations of erythromycin and tylosin. more...
