HIV envelope protein gp120 effect on activated B cells from healthy donors: time course

Temporal analysis of B cells isolated from healthy donors in response to activation in the presence of HIV-1 envelope protein gp120 with high (gp120++) or low (gp120-) affinity for integrin α4β7. Results provide insight into mechanisms by which α4β7-gp120 interaction negatively affects B cells.

HIV-1 gp120 impairs B cell proliferation by inducing TGF-β1 production and FcRL4 expression via an α4β7-dependent mechanism

(Submitter supplied) The anti-HIV humoral immune response following acute infection is delayed and ineffective. HIV envelope protein gp120 binds to and signals through α4β7 on T cells. We show that gp120 also binds and signals through α4β7 on B cells, resulting in an abortive proliferative response. In primary B cells, gp120 signaling through α4β7 resulted in increased expression of TGF-β1 and the B cell inhibitory receptor FcRL4. more...

Characterization of human Fcrl4-positive B cells

(Submitter supplied) FCRL4 is an immunoregulatory receptor that belongs to the Fc receptor-like (FCRL) family. In healthy individuals, this protein is specifically expressed by memory B cells (MBCs) and is preferentially localized in subephitelial regions of lymphoid tissues. An expansion of FCRL4+ B cells has been shown in blood or other tissues in various infectious or autoimmune pathologies. In the present work, we generated and characterized in vitro FCRL4+ B cells from purified MBCs using T-dependent and/or T-independent stimulation. more...
