CD34+derived erythoblast co-culture with Plasmodium falciparum 3D7 (GenePattern analysis)

Analysis of growth factor-mobilized, CD34+ hematopoietic stem cells in vitro differentiated to day 10 (polychromatophilic stage) or day 14 (orthochromatic stage) then infected with P. falciparum for 24 hr. Results provide insight into host erythroblast cell response to infection by P. falciparum.

Human CD34+-derived erythoblast (polychromatophilic and orthochromatic) response to co-culture with Plasmodium falciparum 3D7

(Submitter supplied) Global, genomic responses of erythrocytes to infectious agents have been difficult to measure, because these cells are e-nucleated. We have previously demonstrated that in vitro matured, nucleated erythroblast cells at the orthochromatic stage can be efficiently infected by the human malaria parasite Plasmodium falciparum. We now show that infection of orthochromatic cells induces change in 609 host genes. more...

Transcriptional profiling of primary human erythroblasts infected with Plasmodium falciparum

(Submitter supplied) In malaria infection, Plasmodium spp. parasites accumulate in the bone marrow near sites of erythroid development. While it has been observed that Plasmodium falciparum infection of late-stage erythroblasts can delay terminal erythroid differentiation and enucleation, the mechanism(s) underlying this phenomenon are unknown. Here, we apply RNA-seq after fluorescence-activated cell sorting (FACS) of infected erythroblasts to identify transcriptional responses to direct and indirect interaction with P. more...

The molecular consequences of Nix ablation on apoptosis and erythropoiesis

(Submitter supplied) Normal erythropoiesis requires a critical balance between proapoptotic and antipaoptotic pathways. Bcl-xl, an antiapoptotic protein is induced at end-stages of differentiation of erythroid precursors in response to erythropoietin. The details of the proapoptotic pathway and the critical proapoptotic proteins inhibited by Bcl-xl in erythropoiesis are not well understood. We employed gene targeting to ablate Nix, a proapoptotic BH3-domain only Bcl2 family protein, which is known to be transcriptionally induced during erythropoiesis. more...

Proapoptotic BH3-only protein Nix deficiency effect on the spleen

Analysis of spleens of mutants lacking Nix, a proapoptotic BH3-only factor that is transcriptionally induced during erythropoiesis. Erythropoiesis requires a critical balance between proapoptotic and antipaoptotic pathways. Results provide insight into the role of Nix in erythropoiesis.

Effect of Plasmodium falciparum infected erythrocytes on primary human brain microvascular endothelial cell

(Submitter supplied) Cerebral malaria is a severe multifactorial condition associated with the interaction of high numbers of infected erythrocytes to human brain endothelium without invasion into the brain. The result is coma and seizures with death in more than 20% of cases. Because the brain endothelium is at the interface of these processes, we investigated the global gene responses of human brain endothelium after the interaction with Plasmodium falciparum–infected erythrocytes with either high- or low-binding phenotypes. more...
