All-trans-retinoic acid effect on transglutaminase 2-silenced, NB4 model of acute promyelocytic leukemia: time course

Analysis of stable TG2-knockdown NB4 APL cells induced to differentiate in the presence of all-trans-retinoic acid (ATRA) for up to 72 hrs. Silencing of TG2 delays the differentiation process of NB4 cells. Results provide insight into the role of TG2 in the ATRA-induced differentiation of NB4 cells.

Gene expression profiling of ATRA-differentiated wild-type and TG2 knockout NB4 cells

(Submitter supplied) Treatment of acute promyelocytic leukemia (APL) with all-trans-retinoic acid (ATRA) results in terminal differentiation of leukemic cells toward neutrophil granulocytes. Administration of ATRA leads to massive changes in gene expression, including down-regulation of cell proliferation-related genes and induction of genes involved in immune function. One of the most induced genes in APL NB4 cells is transglutaminase 2 (TG2). more...

The role of TFEB in retinoid induced differentiation of NB4 APL leukemic cells (shTFEB)

(Submitter supplied) We report the use of RNAseq to determine the role of the TFEB transcription factor in all-trans retinoic acid induced differentiation of NB4 acute promyelocytic leukemia (APL) cells. We used lentiviral mediated shRNA approaches to functionally deplete TFEB in NB4 cells and compared the transcriptomes of wild type, scramble control shRNA and shTFEB knockdown cells treated with ethanol (control) versus all-trans retinoic acid for 72 hours.

Genome-wide expression profiling of acute promyelocytic leukemia NB4 cells treated with all-trans-retinoic acid

(Submitter supplied) Acute promyelocytic leukemia (APL) is a hematological disease characterized by a balanced reciprocal translocation that leads to the synthesis of the oncogenic fusion protein PML-RARα. APL is mainly managed by a differentiation therapy based on the administration of all-trans retinoic acid (ATRA) and arsenic trioxide (ATO). However, therapy resistance, differentiation syndrome, and relapses require the development of new low-toxicity therapies based on the induction of blasts differentiation. more...

A PU.1 suppressive target gene, metallothionein-1G, inhibits retinoic acid-induced NB4 cell differentiation

(Submitter supplied) We recently revealed that myeloid master regulator PU.1 directly represses metallothionein (MT)-1G through its epigenetic activity, but the functions of MT-1G in myeloid differentiation remain unknown. To clarify this, we established MT-1G-overexpressing acute promyelocytic leukemia NB4 (NB4MTOE) cells, and investigated whether MT-1G functionally contributes to all-trans retinoic acid (ATRA)-induced NB4 cell differentiation. more...

Long Intergenic Non-Coding RNA HOTAIRM1 Regulates Cell Cycle Progression During Myeloid Maturation in NB4 Promyelocytic Leukemia Cells

(Submitter supplied) HOTAIRM1 is a long intergenic non-coding RNA located in the human HOXA gene cluster, with gene expression highly specific for maturing myeloid cells, particularly during all-trans retinoic acid (ATRA) induction of granolopoiesis in NB4, a human t(15;17) acute promyelocytic leukemia (APL) cell line. We sought to assess the impact of HOTAIR knockdown on the global programme of gene expression underlying the granulocytic maturing process in NB4 cells. more...

NB4 cell line untreated and induced to differentiate by a 48h exposure to 1µM ATRA

(Submitter supplied) NB4 is a cell line model of acute promyelocytic leukemia (APL).The t(15;17) translocation and consequent expression of the PML (Promyelocytic Leukemia Protein)/RARalpha (Retinoic Acid Receptor Alpha) fusion protein blocks differentiation at the promyelocytic stage. The PML portion of the PML/RARalpha fusion protein has a high affinity for the corepression complex, preventing RARalpha transcriptional activity. more...

Combination of triciribine and p38 inhibitors enhances the differentiation effect on myeloid leukemia cells

(Submitter supplied) Differentiation therapy with all-trans retinoic acid (ATRA) is well established for acute promyelocytic leukemia (APL). However, the narrow application and tolerance development of ATRA remain to be improved. A number of kinase inhibitors have been reported to induce differentiation. In this study, we challenged several combinations of these kinase inhibitors. As a result, we found that the combination of Akt inhibitor Triciribine (TCN) and several p38 MAPK inhibitors have profound effect for differentiation in several myeloid cell lines. more...

SALL4 ChIP-chip using NimbleGen HG18 RefSeq Promoter tiling array (2.7kb)

(Submitter supplied) Increasing studies suggest that SALL4 may play vital roles in leukemogenesis. We have used chromatin immunoprecipitation followed by microarray hybridization as a screening tool to determine potential genes that may account for the role SALL4 plays in leukemogenesis. Analysis of SALL4 binding sites reveals that genes involved in cell death, cancer, DNA replication/repair, and cell cycle were highly enriched (p<0.05). more...

Transcriptional Regulation of HL60 Neutrophil Differentiation: Promyelocyte trajectory vs. Neutrophil trajectory

(Submitter supplied) Using the HL60 multipotent promyelocytic leukemia cell line, we performed experiments that lead to two different cell fate attractors by treatments of varying dosage and duration of the differentiation agent all-trans-retinoic acid (ATRA). The dosage and duration combinations corresponding to the two treatments were chosen by means of of cytometric measurements of CD11b, a well-known early differentiation marker, such that the two populations are poised at the same stage of differentiation. more...
