CEA_SGF:E00020#Dose_Effect_Blood

(Submitter supplied) In this study, we asked whether expression profiling could generate a “signature” of a dose of exposure to a toxic in general and to gamma rays in particular. To characterize genes that could potentially “sign” an exposure to irradiation and could allow the estimation of the received dose, we have performed DNA microarray experiments. Responses to gamma rays were analysed by genome-wide expression profiling at different doses in blood leucocytes of c57bl/6 mice. more...

Specific, Durable and Accurate Gene Expression Signatures of Radiation Response in Mice and Humans

(Submitter supplied) Previous work has demonstrated the potential for peripheral blood (PB) gene expression profiling for the detection of disease or environmental exposures. We have sought to determine the impact of several variables on the PB gene expression profile of an environmental exposure, ionizing radiation, and to determine the specificity of the PB signature of radiation versus other genotoxic stresses. Keywords: peripheral blood, gene expression study, radiation reponse

Gene expression profiles from heart, lung and liver samples of total body irradiated minipigs

(Submitter supplied) The present work identified gene expression changes in response to total body irradiation (TBI) in heart, lungs, and liver tissue in Gottingen minipigs. Animals were exposed to 1.7 Gy, 1.9 Gy, 2.1 Gy and 2.3 Gy of TBI and were followed for 45 days. Organ samples were collected at the end of 45 days or sooner if the animal displayed morbidity necessitating euthanasia. RNA was extracted and gene expression profiling studied using Agilent arrays.

A translatable predictor of human radiation injury

(Submitter supplied) The development of a biodosimetric assay based on the combined gene expression analysis of ex vivo-irradiated human peripheral blood (PB) and humans treated with total body irradiation has been developed. Gene expression profile signature of human radiation injury into a rapid and high-throughput chemical ligation-dependent probe amplification assay (CLPA) has been shown to discriminate 6 radiation dose levels in human PB. more...

Dose- and time- dependent ionizing ratidation effect on mice peripheral blood

(Submitter supplied) Gene expression profiles of peripheral blood samples from C57BL/6 mice exposed with ionizing radiation. We used mice as model animal to study biologial recovery response after radiation damage. Therefore, we obtained gene expression profiles from C57BL/6 mice exposed with various levels of ionizing radiation, including low and high doses and control groups. In order to measure recovery rate, we collected peripheral blood samples at different time durations after the exposure. more...

Prediction of in vivo radiation dose in radiotherapy patients using ex vivo and in vivo gene expression signatures

(Submitter supplied) After defining a gene expression signature that predicted radiation exposure dose with high accuracy in human peripheral white blood cells irradiated ex vivo, we now demonstrate the predictive power of gene expression signatures in blood from patients undergoing total body irradiation. Using whole genome microarray analysis, we have identified genes that respond to radiation exposure in cancer patients in vivo. more...

Radiation responses in peripheral white blood cells of smokers and non-smokers

(Submitter supplied) Understanding the possible impact of potential confounding factors is necessary for any approach to radiation biodosimetry. Potential confounding factors have not been fully addressed for gene expression-based biodosimetry approaches, such as we are developing. To begin addressing this need, we have used an ex vivo irradiated peripheral blood cell model to investigate the potential effect of smoking on the global radiation gene expression response, and looked for genes that respond to radiation differently in smokers and non-smokers, and also in males and females. more...

Effect of total-body Irradiation on gene expression in mouse whole blood

(Submitter supplied) For victims of radiological accidents, rapid dose estimation and damage prediction are essential. Administering the gold-standard biodosimetry chromosome aberration assay requires highly skilled individuals and several days of labor; consequently, rapid turnaround is an important concern. Identification of new dose estimation markers and damage-predicting in vivo molecules to replace the chromosome aberration assay is crucial to improving the delivery time of medical treatment. more...

Gene expression in mouse blood following low dose-rate or acute x-ray exposure

(Submitter supplied) Radiation biodosimetry can play a critical role in the response to a large-scale radiologic emergency, and gene expression profiles have shown promise for providing biodosimetric information. This study was designed to test if gene expression could be used to distinguish between doses received from acute exposures and more protracted exposures, such as those that would result from fallout. Mice were exposed to whole body X-rays at low dose rate (LDR, 3.09 mGy/min) for 6, 12, or 24 hours (1.1, 2.2, or 4.4 Gy), or to equivalent doses delivered at high dose rate (HDR, 1.03 Gy/min). more...

Analysis of gene co-expression networks in skin cells exposed to different doses of ionising radiation at different time points

(Submitter supplied) Most studies have analysed the effects of high dose radiation such as atomic bomb survivors in Japan, people exposed during the Chernobyl nuclear accident, patients undergoing radiation therapy, uranium miners, etc. However, it has been difficult to measure and assess the risk of cancer in people exposed to lower doses of ionising radiation, such as the people living at high altitudes, who are exposed to more natural background radiation from cosmic rays than people at sea level. more...

Global gene expression responses to low doses of protons in a human 3-dimensional skin model

(Submitter supplied) Accumulating data suggest that the biological responses to high and low doses of radiation are qualitatively different, necessitating the direct study of low dose responses. Most such studies have utilized 2-dimensional culture systems, which may not fully represent responses in 3-dimensional tissues. To gain insight into low dose responses in tissue, we have profiled global gene expression in EPI-200, a 3-dimensional tissue model that imitates the structure and function of human epidermis, at 4, 16 and 24 hours after exposure to high (2.5 Gy) and low (0.1 Gy) doses of low LET protons. more...
