Timecourse of mCMV Infection of Bone Marrow-Derived Macrophages (BMDMs)

(Submitter supplied) The systematic temporal gene expression analysis of primary macrophages activated under immune (interferon-gamma) and productive viral infection with murine cytomegalovirus (mCMV). The primary objective of the study is to define, in an unbiased manner, cause-and-effect relationships in the program of gene activation in this cellular system. RNA-seq of 8hr LPS-stimulated bone marrow-derived macrophages from wild type, Ch25h-deficient, and LXRab knockout mice

(Submitter supplied) 25-hydroxycholesterol has been demonstrated to regulate SREBP processing, yet Ch25h-deficient mice have no cholesterol abnormalities. Using RNA-seq, we find that LPS-stimulated, Ch25h-deficient BMDMs have dysregulated SREBP target genes, demonstrating that 25-hydroxycholesterol is an induced repressor of SREBP during inflammatory settings.

Transcriptomics analysis of cells transfected with miR-183 cluster mimics and immunostimulated with poly(I:C)

(Submitter supplied) Our work demonstrated that miR-183 cluster regulates IFN production and signaling We used microarrays to detail the effect of miR-183 cluster on hepatic gene expression during immunostimulation with poly(I:C). Our finding suggests enrichment of pathways involved in IFN production and signaling. Amoung the miR-183 repressed genes, we identifies PPP2CA and TRIM27, which are negative regulators of the innate immune response.
