Cumulus-oocyte complex temporal expression

(Submitter supplied) Cumulus-oocyte complexes were isolated a seperate time-points to generate temporal complexes. Targets from two biological replicates at each time point (0h, 8h, 16h post-hCG treatment) were generated and the expression profiles were determined using Affymetrix GeneChip Mouse Genome 430 2.0 Arrays. Comparisons between the sample groups allow the identification of genes with temporal expression patterns. more...

Cumulus oocyte complex during ovulation: time course

Analysis of cumulus oocyte complexes (COCs) isolated from ovaries prior to ovulation or from the oviduct after ovulation. Follicular growth stimulated using equine chorionic gonadotropin (CG) and ovulation induced using human CG. Results identify candidate genes in COCs that impact ovulation.

Gene expression analysis of pig cumulus-oocyte complexes stimulated in vitro with follicle stimulating hormone or epidermal growth factor-like peptides

(Submitter supplied) Background: The gonadotropin-induced resumption of oocyte meiosis in preovulatory follicles is preceded by expression of epidermal growth factor (EGF)-like peptides, amphiregulin (AREG) and epiregulin (EREG), in mural granulosa and cumulus cells. Both the gonadotropins and the EGF-like peptides possess the capacity to stimulate resumption of oocyte meiosis in vitro via activation of a broad signaling network in cumulus cells. more...

Estrus Synchronization and Ovarian Hyper Stimulation Treatments Have Negligible Effects on Cumulus Oocyte Complexes Gene Expression: Induction of Ovulation Causes Major Expression Changes

(Submitter supplied) The effects of exogenous hormones used for estrus synchronization, and ovarian hyper stimulation on cumulus oocyte complexes (COCs) gene expression in sexually mature rats were determined using microarrays. Gene expression in COCs collected from GnRH (Gtrt), GnRH+eCG (G+Etrt), and GnRH+eCG+hCG (G+E+Htrt) treatments were compared to COCs from naturally cycling (NC) rats. There was no significant difference in gene expression among NC, Gtrt and G+Etrt. more...

Preovulatory ovary

(Submitter supplied) Nylon microarrays displaying 9152 rainbow trout cDNAs were hybridized using RNA samples originating from ovarian tissue collected during late vitellogenesis, post-vitellogenesis and oocyte maturation. Differentially expressed genes were identified using a statistical analysis. A supervised clustering analysis was performed using only differentially expressed genes in order to identify gene clusters exhibiting similar expression profiles. more...

Preovulatory ovaries: time course

Analysis of fish preovulatory ovaries collected during late vitellogenesis, post-vitellogenesis and oocyte maturation. In fish, the post-vitellogenic period is critical for completion of the oogenetic process. Results provide insight into molecular events occurring during the preovulatory period.

Comparison of transcriptomes of mouse cumulus cells collected from mice before and after 6-h of hCG priming.

(Submitter supplied) We sought to identify genes that are regulated by the ovulatory signals in mouse cumulus cells. We compared the transcriptomes between cumulus cells colected from mice before (hCG-0h) and after 6 h (hCG-6h) of hCG injection.

Comparison of transcriptomes between DMSO-5uM and Torin1-5uM treated mouse cumulus-oocyte complexes cultured in vitro for 14h

(Submitter supplied) We sought to identify the potential specific roles of the MTOR signalling in cumulus cells by comparing the transcriptomes of the Control (treated with the DMSO vehicle) and MTOR-specific inhibitor Torin 1(5uM)-treated cumulus-oocyte complexes that were cultured for 16 hours. We compared the transcriptomes between DMSO- and Torin 1- treated cumulus-oocyte complexes.

Small RNA profiling of human cumulus cells and oocytes

(Submitter supplied) Cumulus cells are biologically distinct from other follicular cells and perform specialized roles, transmitting signals within the ovary and supporting oocyte maturation during follicular development. The bi-directional communication between the oocyte and the surrounding cumulus cells is crucial for the acquisition of oocyte competence. Using Illumina/deep-sequencing technology, we dissected the small RNAome of pooled human mature MII oocytes and cumulus cells.

Gene expression analysis of granulosa cells from Fzd1 WT and KO mice

(Submitter supplied) WNT4 is required for normal ovarian follicle development and female fertility in mice, but how its signal is transduced remains unknown. Fzd1 encodes a WNT receptor whose expression is markedly induced in both mural granulosa cells and cumulus cells during the preovulatory period, in a manner similar to Wnt4. To study the physiological roles of FZD1 in ovarian physiology and to determine if it serves as receptor for WNT4, Fzd1-null mice were created by gene targeting. more...

Integrated analysis of transcriptome and histone modifications in granulosa cells during ovulation in female mice [RNA-seq]

(Submitter supplied) The ovulatory LH surge induces rapid changes of transcriptome and cellular functions in granulosa cells (GCs) undergoing luteinization. However, it remains unclear how the changes in genome-wide gene expression are regulated. The changes of trimethylation of lysine 4 on histone H3 (H3K4me3) is associated with the rapid alteration of gene expression. In this study, we genome-widely investigated the changes of transcriptome and H3K4me3 status in mouse GCs undergoing luteinization during ovulation. more...
