TAZ-mediated enhancement of adipogenesis in goat adipose mesenchymal stem cells induced by TNF-α

(Submitter supplied) Obesity is a clinical condition characterised by a chronic, low-grade inflammatory state. Adipose-derived mesenchymal stem cells (ADSCs) have a multidirectional differentiation potential to adipocytes. However, the relationship between adipogenesis and inflammation is not yet fully understood.TAZ (WW domain containing transcription regulator 1) can act as a molecular rheostat to fine-tune the osteoblast and adipocyte differentiation balance. more...

Identification of LncRNAs/circRNAs associated with testicular development and spermatogenesis in black goat

(Submitter supplied) Establishment and maintenance of spermatogenesis need a complex process and vast regulatory network. There is growing evidence reveals that long noncoding RNAs (lncRNA) plays important role in regulating testicular development and spermatogenesis in a stage-specific way. We here report the identification of lncRNA LOC108635509 as key lncRNA regulator in black goat spermatogenesis. In the current study, we screened the transcriptomes (lncRNA and mRNA) of testicular from Guangxi black goats before puberty (3 days old, D3; 30 days old, D30), puberty (90 days old, D90) and postpuberty (180 days old, D180), and found there were 1211, 12180, 834 differential lncRNAs and 1196, 8838,269 differential mRNAs at the ages of D30 vs D3, D90 vs D30, and D180 vs D90. more...

Transcriptome analysis reveals miRNA expression profiles in hypothalamus tissues during the sexual development of Jining grey goats

(Submitter supplied) In order to reveal the changes of microRNA spectrum in hypothalamus tissues of goats from birth to sexual maturity, smallRNA sequencing was performed on hypothalamus tissues of Jining grey goats at 4 developmental stages after birth. Twenty libraries from 4 different developmental stages (5 goats per stage) were successfully constructed and the corresponding miRNA expression profiles were obtained.

Combined analysis of temporal metabolomics and transcriptomics reveals the metabolic patterns in goat oocytes during maturation

(Submitter supplied) Well-balanced and orderly metabolism is a crucial prerequisite for promoting oogenesis. Involvement of single metabolites in oocyte development has been widely reported; however, the comprehensive metabolic framework controlling oocyte maturation is still lacking. In the present study, we employed an integrated temporal metabolomic and transcriptomic method to analyze metabolism in goat oocytes at key stages, revealing the global picture of the metabolic patterns during maturation. more...

Compartmentalisation proteomics revealed endolysosomal protein network changes in goat model of atrial fibrillation

(Submitter supplied) To identify the role of the endolysosomal (EL) protein network in atrial fibrillation (AF), we applied a newly developed organelle isolation method to the AF goat model. To accomplish the goal, we used proteomics, transcriptomics, integrated analysis, enzyme assays, electron tomography, and western blotting and identified several regulating EL proteins, related protein networks and pathways.

Direct Reprogramming of Fibroblasts into Functional Mammary Epithelial cells by small molecules (bulk miRNA-seq)

(Submitter supplied) Mammary epithelial cells (MECs), the only cell type which could be capable of secreting milk in mammary gland, can be applied for the generation of transgenic mammary cell/gland bioreactor. However, continuously achieving large amount of high-quality goat MECs by non-invasion method, especially prepuberal MECs, in vitro has been a challenge. Here, we show that a small molecules cocktail enables the in vitro generation of chemically-induced mammary epithelial cells (CiMECs) with milk-secreting properties from goat ear fibroblasts. more...

Single-Nucleus Transcriptomic Atlas of Goat (Capra hircus) Ovarian Prolificacy II

(Submitter supplied) Molecular mechanisms of follicular atresia and prolificacy of mammal remain unclear. We surveyed the single-cell transcriptomic landscape of ovaries from single and prolificacy goat and identified seven ovarian cell types with distinct gene-expression, transcriptional factor networks and reciprocal interactions signatures. In-depth dissection of gene-expression dynamics of granulosa cells (GCs) that displayed development stage-specific expression patterns and specific gene signatures were identified that may reflect developmental competency and ovarian reserve. more...

Divergent composition and transposon-silencing activity of small RNAs in mammalian oocytes

(Submitter supplied) We found piRNAs with different lengths represented the predominant small RNA species in oocytes from the 12 explored species, except mouse. We found endo-siRNAs resulted from the truncated Dicer isoform were mouse-specific, and os-piRNAs associating with PIWIL3 in human oocytes are widespread in mammals and are typically with low levels of the 2’-3’-O-methylation. The sequences of many highly expressed piRNA clusters are fast-evolving compared with their syntenic genomic locations, and the TE families distributing in the conserved piRNA clusters are various between species.

Integrating into dairy goats chromatin accessible sites during lactation promotes transgene expression efficiency

(Submitter supplied) Combining RNA-seq with ATAC-seq, we screened the open chromatin of dairy goat mammary epithelial cells during lactation and then integrated human lysozyme into various open regions using the CRISPR/Cas9 system. A comparison of lysozyme gene expression and integration efficiency showed that chromatin accessibility at target sites was significantly positively correlated with gene insertion efficiency, and the insertion of foreign genes into highly accessible chromatin regions could enhance its expression.

Milk fat miRNome changes in response to LPS challenge in Holstein cows

(Submitter supplied) Cows were fed a lactation diet at ad libitum intake (n = 6). At 27±3 days in milk, cows were injected with 50 µg of LPS E. coli in one healthy rear mammary quarter. Milk samples were collected just before LPS challenge (LPS-) and 6.5 h after LPS challenge (LPS+) from the same cows. Microarray analysis was performed using customized 8x60K ruminant miRNA microarrays to compare LPS- to LPS+ miRNome. MiRNome comparison between LPS- and LPS+ identified 37 differentially abundant miRNAs (q-value ≤ 0.05)

Single-cell transcriptome analysis of the germ cells and somatic cells during mitotic quiescence stage in goats

(Submitter supplied) As goats are important domestic animals, the regulation of germline and gonadal somatic cells in goats during the mitotic quiescence phase has not been investigated previously. Characterization of microRNA profiles in the mammary gland tissue of dairy goats at the late lactation, dry period and late gestation stages

(Submitter supplied) The mammary gland redeveloped to the pre-pregnancy state during involution, which shows that the mammary cells have the characteristics of remodeling. The rapidity and degree of mammary gland involution are different between mice and dairy livestock (dairy cows and dairy goats). However, the molecular genetic mechanism of miRNA in involution and remodeling of goat mammary gland has not yet been clarified. more...
