Cuttings of hybrid aspen (Populus tremula x tremuloides) clone T89 (wild type) and the transgenic lines were grown in half-strength Murashige/Skoog medium (½ MS) under sterile conditions for approximately 4 weeks and then used for In vitro rooting experiments.

Extraction protocol |
RNA was extracted using the RNeasy micro kit (QIAGEN). We added a first clean-up step with QIAshredder Mini Spin Columns (QIAGEN) and an on-column DNA digestion step with DNAse I (Qiagen). RNA quality was verified by Experion StdSens capillary gels (Bio-Rad). Total RNA preparations were amplified using the SMARTer PCR cDNA Synthesis Kit (Clontech) according to the manufacturer’s instructions.
