Title |
HepG2_Stearic acid_rep2 |
Sample type |
RNA |
Source name |
HepG2, Stearic acid, 7 days
Organism |
Homo sapiens |
Characteristics |
cell line: HepG2 incubated fatty acid: Stearic acid
Treatment protocol |
Incubation for seven days with 100 µM fatty acid (oleic, elaidic or stearic acid)
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA Purification Kit (NORGEN BIOTEK CORPORATION) Distribubed by Phalanx Biotech
Label |
Cy5
Label protocol |
RNA was converted to double-stranded cDNA and amplified using an in vitro transcription reaction that included aminoallyl-UTP, and the aRNA product was conjugated with Cy5T NHS ester
Hybridization protocol |
Fragmented aRNA was hybridized at 42° C overnight using the HybBag mixing system with 1× OneArray Hybridization Buffer (Phalanx Biotech), 0.01 mg/mL sheared salmon sperm DNA (Promega) and 0.025 mg/mL labeled target. After hybridization, the arrays were washed according to the OneArray protocol
Scan protocol |
Raw intensity signals from each microarray were captured using a Molecular Dynamics™ Axon 4100A scanner, measured using GenePixPro™ Software, and stored in GPR format
Description |
SB raw data file technical rep1: H027-0607002924.gpr raw data file technical rep2: H028-0607002925.gpr
Data processing |
The data from all microarrays in each experimental set were passed to Rosetta Resolver (Rosetta Biosoftware) for analysis. Testing was performed by combining the technical replicates and performing statistical analyses using Rosetta Resolver's proprietary modeling techniques
