DU145 prostate cancer cell line was obtained from American Type Culture Collection. Cells were maintained in RPMI 1640 supplemented with 10% heat-inactivated FBS. Cell cultures were maintained at 37°C under a humidified atmosphere of 5% CO2 in an incubator.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip HG-U133A 2.0 arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

GeneChips were scanned using Affymetrix GeneChip Scanner 3000 7G.
