Total RNA was extracted from monkey tonsillar throat swabs with the FastPrep FP 120 kit (Qbiogene, Carlsbad, CA) by immersing the swab tips directly into the FastPrep Blue tubes containing 300 ul 5 mM ammonium aurintricarboxylate, 500 ul CRSR-Blue(Qbiogene), and acid phenol:chloroform (5:1, pH 4.5; Ambion, Austin, TX). The mixture was homogenized (80 sec at speed #5), heated at 65 oC for 20 min, and centrifuged at 16,000xg for 15 min. The aqueous phase was collected, 250 ug glycogen (Roche Applied Science, Indianapolis, IN) and 0.8 ug MS2 carrier RNA (Roche Applied Science, Indianapolis, IN) was added, and the mixture was concentrated to 100 ul with a vacuum concentrator (Eppendorf, Germany). The concentrate was purified using the Qiagen RNeasy 96 kit following the manufacturer's protocol (Qiagen, Valencia, CA).
