Treatment protocol PAECs were treated with with vehicle control (DMSO, Sigma-41639), BRD2889 (1uM) (Broad Institute), as indicated. After exposure to hypoxia for 24 h, the cells were harvested for assay.
Growth protocol Pulmonary arterial endothelial cells (PAECs) plated in 6-well cell culture plates at 1X 10^5 cells per well, allowed to grow for 24 h, and the media were then changed to standard media and then the cells were placed into the hypoxia chamber.
Extracted molecule total RNA
Extraction protocol RNeasy kit (Qiagen) using extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol RNA samples were labeled using the Affymetrix WTPLus kit according to manufacturer’s guidelines.
 
Hybridization protocol RNA samples were probed using the Clariom S Human Array.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000 7G System.
Description HPAECs under normoxia and treated with vehicle
Data processing The gene expression was normalized with Robust Multiarray Analysis (RMA). The differential expression of the genes was determined using Affymetrix transcriptome analysis console (TAC) software.
