Treatment protocol Pregnant females were subcutaneously injected twice with 0.4mg/kg of dexamethasone at embryonic stages 17.5 and 18.5. (Only in SAMPLES 15, 16 and 17)
Growth protocol Animals were housed in type IIL individually ventilated cages, in a temperature- and humidity-controlled room with a 12h light/dark cycle, with food (Teklad 2014, Harlan Laboratories) and water available ad libitum. All the lung samples were collected from postnatal day 0 mice.
Extracted molecule total RNA
Extraction protocol Manually selected lungs of Control, HRas-KO, NRas-KO and DKO from postnatal day 0 were used for RNA extraction using the TRIZOL protocol. RNA was further purified using Qiagen Rneasy mini kit. Each RNA sample for each microarray was obtained from one individual animal.
Label biotin
Label protocol Samples were enzymatically fragmented and biotinylated using the WT Terminal Labeling Kit (Ambion)
 
Hybridization protocol Samples were hybridized using Affymetrix hybridization kit materials • Washing and scanning were performed using GeneChip System of Affymetrix (GeneChip Hybridization Oven 640, GeneChip Fluidics Station 450)
Scan protocol Affymetrix Gene ChIP Scanner 3000 7G
Description Gene expression data from whole HRAS and NRAS-deficient lung extracts treated antenatally with dexamethasone.

Data processing The RMA function provided by the package Affy from R/Bioconductor was applied to the raw expression data.
