Growth protocol |
Spleens from C57BL/6 male mice were collected at 12 weeks of age and mechanically smashed by plunger of a 2ml syringe with a 40 μm cell strainer. Splenocytes were enriched for CD8+ T cells with the MojoSort mouse CD8+ naïve T cell isolation kit and stained for FACS sorting. FACS-sorted cells were lysed with 50 mL RLT buffer, with the addition of 1 mL RNAse inhibitor
