Treatment protocol |
Mice were transplanted with primary leukemias. Ten days later mice were treated intraperitoneal (ip) with 1mg tamoxifen or vehicle for 5 days. Afterwards, mice were treated ip with 100 mg/Kg cytarabine for 5 days and 3mg/kg doxorubicin for 3 days or PBS as control.

c-Kit+ bone marrow cells (isolated with magnetic beads) from Hif-1alphaLox/Lox; CreERT2 mice were transduced with MLL-AF9-retrovirus and transplanted into wild type mice to generate primary leukemic cells.

3 days after end of treatment, bones (crista, femur, tibia) were collected from mice and bone marrow extracted by crushing bones in mortar. GFP+ cells (MLL-AF9-transduced) were sorted by flow cytometry on a BDFacsAriaII using 70 micron nozzle. Libraries were constructed according to 10x genomics Chromium Single Cell 3' Reagents Kit V2 Protocol
