Treatment protocol |
3T3-L1 pre-adipocytes were seeded at 90,000 cells per well in a 12-well plate, followed by transfection with siRNA (control or against 14-3-3ζ). 24 hr after transfection, cells were induced to differentiate with of 172 nM insulin, 500 μM IBMX, and 500 nM dexamethasone in differentiation media containing 25 mM glucose DMEM and 10% fetal bovine serum.

Growth protocol |
3T3-L1 pre-adipocytes were grown in 25mM glucose DMEM, supplemented with 10% newborne calf serum.

Extraction protocol |
Total RNA was extracted using the Qiagen Rneasy kit with the on-column Dnase treatment. Samples were stored at -80 C until use. Libraries were generated using Illumina's Truseq V2 library kit, and each sample was given a unique identifier prior to sequencing on an Illumina HiSeq 2500 (150 bpx2, 20 million paired-end reads)
