Extraction protocol Proximal tibia was removed, flash frozen in liquid nitrogen, cryosectioned and transferred to adhesive membrane. SFZ and deepAC were separately collected by LMD. Total RNA was extracted using Picopure RNA Isolation Kit. Amplified RNA (aRNA) was generated by linear amplification of mRNA using RiboAmp HS PLUS Kit. Illumina TruSeq RNA Sample Prep Kit v2 was used with 50ng of aRNA for the construction of sequencing libraries.
RNA libraries were prepared for sequencing using standard Illumina protocols
