Title |
Patient 8 - skin-week 7 visit |
Sample type |
RNA |
Source name |
human skin
Organism |
Homo sapiens |
Characteristics |
treatment: fresolimumab two doses 1 mg/kg at baseline and 3 wks
Extracted molecule |
total RNA |
Extraction protocol |
Skin biopseie placed in RNAlater at bedside were stored at -80 degrees C until processing. RNA was exrtacted by homogenization in RNAzol and prifying over Rneasy columns
Label |
biotin
Label protocol |
Biotin labeling was performed using the 3' IVT Express Kit (Affymetrix, Santa Clara, CA) according to the manufacturer's protocol.
Hybridization protocol |
The labeled, fragmented DNA was hybridized to the GeneChip HG-U133A 2.0 Array (Affymetrix, Santa Clara, CA) for 18 hours in a GeneChip Hybridization oven 640 at 45oC with rotation (60 rpm). The hybridized samples were washed and stained using an Affymetrix fluidics station 450
Scan protocol |
After staining, microarrays were immediately scanned using an Affymetrix GeneArray Scanner 3000 7G Plus (Affymetrix, Santa Clara, CA).
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 500.
