Treatment protocol |
Cholesterol deprivation were started when worms hatched. Worms were exposed to fasting for 48 hr since day 2 adult.

Synchronized were grown on NGM plates until young adult and then transferred to FUDR-containig plates.

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100 ng total RNA.

Hybridization protocol |
Following fragmentation, 12.5 ug of cRNA were hybridized for 16 hr at 45C on GeneChip C. elegans Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station.

Scan protocol |
GeneChips were scanned using an Affymetrix GeneChip Scanner.

Data processing |
The data were analyzed with GeneSpring 12.5 GX. Expression signals of probe sets were calculated using RMA algorithm.
