Title The establishment of the anther somatic niche with single cell sequencing
Organism Zea mays
Experiment type Expression profiling by high throughput sequencing
Summary Maize anthers ranging in size from 0.5mm to 3.0mm in increments of 0.25mm were dissected and their cells isolated using the FX-Cell protocol. scRNA-seq libraries were prepped using the CEL-Seq2 protocol and sequenced with 150bp PE reads with Illumina HiSeq or NovoSeq. The developmental trajectories of the somatic cell layers were investigated.

Overall design Anthers were dissected from maize plants of the fertile inbred line W23. Anthers ranging in size from 0.5mm to 3.0mm in increments of 0.25mm were dissected, isolated using a Hana Cell Sorter (NamoCell) and used for scRNA-seq. At least two separate W23 plants provided anthers for each stage
