Title |
The establishment of the anther somatic niche with single cell sequencing |
Organism |
Zea mays |
Experiment type |
Expression profiling by high throughput sequencing
|
Summary |
Maize anthers ranging in size from 0.5mm to 3.0mm in increments of 0.25mm were dissected and their cells isolated using the FX-Cell protocol. scRNA-seq libraries were prepped using the CEL-Seq2 protocol and sequenced with 150bp PE reads with Illumina HiSeq or NovoSeq. The developmental trajectories of the somatic cell layers were investigated.

Overall design |
Anthers were dissected from maize plants of the fertile inbred line W23. Anthers ranging in size from 0.5mm to 3.0mm in increments of 0.25mm were dissected, isolated using a Hana Cell Sorter (NamoCell) and used for scRNA-seq. At least two separate W23 plants provided anthers for each stage
