Gene Expression Profile during human CD4+ T cell differentiation

(Submitter supplied) Subpopulations of human fetal thymocyte and circulating naïve T cells were obtained through FACS sorting, including CD3-CD4+CD8- intrathymic T progenitor cells (ITTP), CD3intCD4+CD8+ "double positive" thymocytes (DP), CD3highCD4+CD8- "single positive" thymocytes (SP4), CD3+CD4+CD8-CD45RA+CD62L+ naïve T cells from cord blood (CB4+), and CD3+CD4+CD8-CD45RA+CD62L+ naïve T cells from adult blood (AB4+). more...

Sensitive expression profiling of T-cell subsets reveals parallel post-thymic differentiation for CD4+ and CD8+ lineages

(Submitter supplied) Gene expression profiling has opened new directions of investigation in scientific research, however, the need for substantial amount of biological material to perform these experiments limits significantly its application range. Here, we have developed procedures to perform microarray analysis on amplified cDNA from single cells, including primary T lymphocytes, and applied this technology to the study of CD4 and CD8 lineage differentiation. more...

Transcriptome profiling of human thymic and peripheral blood CD4 + and CD8+ T cells, using RNA-seq

(Submitter supplied) We have performed high-throughput RNA sequencing to characterize the transcriptomes of primary single positive (SP) CD4+ and CD8+ T cells from infant thymic tissue, as well as primary CD4+ and CD8+ T cells from infant and adult peripheral blood, to enable the comparisons across tissues and ages

Transcriptome analysis of murine thymocytes reveals age-associated changes in thymic gene expression

(Submitter supplied) The decline in adaptive immunity, naïve T-cell output and a contraction in the peripheral T cell receptor (TCR) repertoire with age are largely attributable to thymic involution and the loss of critical cytokines and hormones within the thymic microenvironment. To assess the molecular changes associated with this loss of thymic function, we used cDNA microarray analyses to examine the transcriptomes of thymocytes from mice of various ages ranging from very young (1 month) to very old (24 months). more...

Generation of Tumor Antigen-Specific iPSC-Derived Thymic Emigrants (iTE) using a Novel 3D Thymic Culture System

(Submitter supplied) Induced Pluripotent Stem Cell (iPSC)-derived T cells may provide future therapies for cancer patients, but those generated by current methods, such as the OP9/DLL1 system, have shown abnormalities that pose major barriers for clinical translation. Our data indicate that these iPSC-derived CD8 single positive T cells are more similar to CD4+CD8+ double positive T cells than to mature naïve T cells, display markers of developmental arrest and exhibit characteristics of innate-like immune cells. more...

Expression data from T cells in vivo

(Submitter supplied) Induced pluripotent stem cells (iPSC) might provide an unlimited source of T cells for a variety of clinical applications, including restoration of T cell immunity following stem cell transplantation and generation of minimally differentiated antigen-specific T cells for adoptive immunotherapy. However, current methods of differentiating T cells from iPSC using OP9/DLL1, an extrinsic source of Notch signaling, result in the accumulation of unconventional T cells either lacking CD8 co-receptors or expressing CD8αα homodimers. more...
