Liver response to perinatal exposure to 2,2',4,4'-tetrabromodiphenyl ether: dose response

Analysis of livers of animals exposed to low doses of 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) from gestational day 15 to post-natal day 20. Polybrominated diphenyl ethers are flame-retardant chemicals used in consumer products. Results provide insight into the mechanisms of toxicity of BDE-47.

Liver of rat offsprings exposed perinatally to low doses of 2,2',4,4'-tetrabromodiphenyl ether

(Submitter supplied) Background: Polybrominated diphenyl ethers are a group of flame-retardant chemicals appearing increasingly in the environment. Their health effects and mechanisms of toxicity are poorly understood. Objectives: We screened for the sensitive effects and mechanisms of toxicity of 2,2 ,4,4 -tetra-bromodiphenyl ether (BDE-47) by analyzing the gene expression profile in rats exposed to doses comparable to human exposure. Methods: Wistar dams were exposed to vehicle or BDE-47 (0.002 and 0.2 mg/kg body weight) every fifth day from gestation day 15 to postnatal day 20 by injections to caudal vein. more...

Expression data from PBDE treated rats at PND22 and Week 13

(Submitter supplied) Liver gene transcripts patterns were used to characterize toxicity from exposure to polybrominated diphenyl ethers (PBDEs), flame retardant components. In this study, Wistar Han dams were exposed by gavage to the PBDE mixture (DE71) starting at gestation day 6 (GD 6) and continuing to weaning on postnatal day 21 (PND 21). Offspring from the dams began PBDE direct dosing on PND 12 and were dosed daily through PND 21. more...

Polybrominated diphenyl ether effect on the liver

Analysis of livers collected from postnatal day (PND) 22 and 13-week-old Wistar/Han rats exposed in utero to polybrominated diphenyl ether (PBDE) and directly dosed with PBDE. PBDEs are flame retardants. Results provide insight into the molecular basis of PBDE toxicities.

Transcriptomic profiling of PBDE-exposed HepaRG cells unveils critical lncRNA- PCG pairs involved in intermediary metabolism

(Submitter supplied) Purpose: The goal of this study was to investigate how PBDEs regulate both PCGs and lncRNAs in a PXR/CAR ligand-dependent and independent manner Methods: HepaRG cells, which are human-derived hepatic cells that accurately represent gene expression profiles of human liver tissue, were exposed to BDE-47 and BDE-99 at a dose of 25 μM for 24 hours. Differentially expressed lncRNA-PCG pairs were identified through DESeq2 and HOMER; significant canonical pathways were determined through Ingenuity Pathway Analysis (IPA). more...

Environmentally relevant concentrations of the flame retardant BDE-209 in sediment cause minimal effects on Zebrafish embryos.

(Submitter supplied) Polychlorinated diphenyl ethers are lipophilic, persistent, and bioaccumulable compounds widely used as flame-retardants. These are chemicals of increasing environmental concern due to their lipophilic, persistent, and bioaccumulable characteristics. The objective of this study was to analyze the potential bioavailability and bioaccumulation of BDE-209 as a source of toxicity. Zebrafish embryos were exposed for 8 days to sediments spiked with an environmentally relevant concentration of BDE-209. more...

Developmental Exposure to 2,2’,4, 4’ –Tetrabromodiphenyl Ether Induces Long-Lasting Changes in Liver Metabolism in Male Mice

(Submitter supplied) Polybrominated diphenyl ethers (PBDEs) were used as flame-retardant additives in a wide range of polymers starting in 1965 and were recently withdrawn from commerce in North America and Europe. Generations that were exposed perinatally to the highest environmental doses of PBDE have now reached 5-20 years of age and in the U.S. account for 1/5 of the total population. Emerging data indicates long-term impairment of metabolic health by PBDE exposure in humans and laboratory animals. more...
