Pediatric malignant germ cell tumors: yolk sac tumors and seminomas

Analysis of yolk sac tumors (YSTs) and seminomas, the two principal histological variants of pediatric malignant germ cell tumors (MGCTs). Seminomas and YSTs are presumed to arise from the same progenitor cell. Results provide insight into the molecular basis for the divergence of pediatric MGCTs.

Gene expression analysis comparing between pure- and mixed type testicular seminoma

(Submitter supplied) We hypothesized that the comparison between pure type seminoma and seminoma components in mixed tumor (mixed type seminoma) could reveal early changes of the reprogramming process; however, only a little data is available. We performed gene expression microarray analysis of 6 pure type and 6 mixed type seminomas, and the hierarchical clustering analysis properly grouped each type of seminomas into a separated cluster. more...

Expression Profiling of Adult Male Germ Cell Tumors

(Submitter supplied) Expression profiling of a panel of 101 adult male germ cell tumors and 5 normal testis specimens was performed on Affymetrix U133A and U133B microarrays. This data has been used to: 1) generate a gene classifier that predicts histology (see PMID 15870693). 2) identify candidate target genes on 12p, a region that is gained in almost 100% of germ cell tumors (see PMID 16424014) 3) identify pluripotency associated genes through comparison of pluripotent embryonal carcinoma vs. more...

Genotyping and analysis of chromosome copy number variation (CNV) from pediatric primary intracranial germ cell tumor

(Submitter supplied) Intracranial pediatric germ cell tumors (GCTs) have different histological differentiations, prognosis and clinical behaviors. Prognosis of patients with germinoma and mature teratoma is good, while patients with other types of GCTs, termed as nongerminomatous malignant germ cell tumors (NGMGCTs), require more extensive drug and irradiation treatment regimen. The mechanisms underlying different prognosis of various GCT subgroups remain elusive. more...

Gene Expression Signatures Prognostic for Relapse in Stage I Testicular Germ Cell Tumors (TGCT)

(Submitter supplied) Design, Setting and Participants We reviewed patients with SI non-seminoma (NS) and seminoma (S) from an institutional database (2000-2012) managed with active surveillance. NR-NS and NR-S patients were defined as having no evidence of relapse after 2 and 3 years of surveillance respectively. RNA extraction and gene expression analysis was performed on archival primary tumor samples. Outcome measurements and statistical analysis Gene-Set-Enrichment-Analysis (GSEA) was conducted in order to identify discriminating biological pathways associated with differentiation of R from NR and S from NS. more...

Embryonic stem cells and pluripotent germ cell tumors

(Submitter supplied) Remarkably little is known about the transcriptional profiles of human embryonic stem (ES) cells or the molecular mechanisms that underlie their pluripotency. To identify commonalties among the transcriptional profiles of different human pluripotent cells and to search for clues into the genesis of human germ cell tumors, we compared the expression profiles of human ES cell lines, human germ cell tumor cell lines and tumor samples, somatic cell lines, and testicular tissue samples by using cDNA microarray analysis. more...

Gene expression analysis of human seminoma samples

(Submitter supplied) Purpose: The aim of the present study was to examine the biological differences between seminomas with occult and clinically apparent metastases at the time of diagnosis of the primary tumor to gain insight into the biology of these tumors and facilitate the identification of novel predictors of seminoma metastasis. Materials and Methods: Total RNA including small RNAs was isolated from testicular tumors of patients with pure seminoma presenting with lymphogenic metastasis (n=5, clinical stage IIb/c) and occult metastasis (n=5, clinical stage I). more...

Genomic Landscape, Evolution and Chemoresistance of Yolk Sac Tumors

(Submitter supplied) Yolk sac tumors (YSTs) are a major histological subtype of malignant ovarian germ cell tumors, and compared to other subtypes, patients with YST have a worse prognosis. The molecular basis of this disease has not been characterized at the genomic level. Here we characterized 41 clinical tumor samples (and related normal samples) from 30 YST patients, with distinct responses to cisplatin_x001f__x001F_-based chemotherapy, through a combination of whole-exome and RNA sequencing. more...
