Bone morphogenic protein 6 effect on an endothelial cell line: time course

Analysis of endothelial cells treated for up to 24 hours with bone morphogenic protein 6 (BMP6). BMP proteins participate in the regulation of angiogenesis in physiological and pathological conditions. Results provide insight into the mechanisms that contribute to BMP-dependent angiogenic signaling.

Gene Expression Profile identify a role for cyclooxygenase 2-depedent prostinoid generation in BMP6-induced

(Submitter supplied) The bone morphogenetic protein (BMP) family of proteins participates in an underappreciated signaling pathway involved in regulating angiogensis in physiological and pathological conditions. Microarray profiles of BMP-responsive genes in the endothelium performed by our lab suggest important roles for cyclooxygenase-2 (Cox2) as a potential downstream regulator of BMP-induced angiogenesis. Cox2 is upregulated by BMP6 at both mRNA and protein levels. more...

BMP/Taz-Hippo signaling modulates VEGF-mediated angiogenesis

(Submitter supplied) The BMP/TGFβ-Smad, Notch and VEGF signaling guides formation of endothelial tip and stalk cells. However, the crosstalk of bone morphogenetic proteins (BMPs) and vascular endothelial growth factor receptor 2 (VEGFR2) signaling has remained largely unknown. We demonstrate that BMP family members regulate VEGFR2 and Notch signaling, and act via TAZ-Hippo signaling pathway. BMPs were found to be regulated after VEGF gene transfer in C57/Bl6 mice and in a porcine myocardial ischemia model. more...

Gene expression in aortic roots of wild type versus klotho-deficient mice at 6 weeks of age

(Submitter supplied) Klotho-deficient mice develop aortic valve annulus calcification by 6 weeks of age. Understanding the molecular basis by which aortic valve calcification is initiated will help define potential molecular targets which may be inhibited to reduce or prevent aortic valve calcification. Changes in gene expression related to aortic valve annulus calcification were analyzed by comparing gene expression in the aortic roots from wild type versus klotho-deficient mice.

HDAC5 is a repressor of angiogenesis and determines the angiogenic gene expression pattern of endothelial cells

(Submitter supplied) Class IIa histone deacetylases (HDACs) are signal-responsive regulators of gene expression involved in vascular homeostasis. To investigate the differential role of class IIa HDACs for the regulation of angiogenesis, we used siRNA to specifically suppress the individual HDAC isoenzymes. Among the HDAC isoforms tested, silencing of HDAC5 exhibited a unique pro-angiogenic effect evidenced by increased endothelial cell migration, sprouting and tube formation. more...

RNA-sequencing signatures of liver endothelial cells from iron-adequate and iron-loaded mice

(Submitter supplied) Iron induces hepcidin by activating bone morphogenetic protein (BMP)6-SMAD signaling, which is critical for regulation of systemic iron homeostasis. High iron levels induce BMP6 production in liver endothelial cells, but the molecular mechanisms by which iron regulates BMP6 are incompletely understood. To address this, we performed RNA-sequencing on sorted liver endothelial cells from iron-adequate and iron-loaded mice.

A time series study on BMP6 induced osteoblast differentiation of human mesenchymal stem cells (hMSC)

(Submitter supplied) BMP6 mediated osteoblast differentiation plays a key role in skeletal development and bone disease. Unfortunately, the signaling pathways regulated by BMP6 are largely uncharacterized due to both a lack of data and the complexity of the response. To better characterize the signaling pathways responsive to BMP6, we conducted a time series microarray study to track BMP6 induced osteoblast differentiation and mineralization.
