Gene expression in primate postnatal brain through lifespan - cerebellar cortex

(Submitter supplied) We investigated molecular changes during human, chimpanzee, and rhesus macaque postnatal brain development at the transcriptome, proteome, and metabolome levels in two brain regions: the prefrontal cortex (PFC) that is involved in several human-specific cognitive processes, and the cerebellar cortex (CBC) that may be functionally more conserved. We find a nearly three-fold excess of human-specific gene expression changes in PFC compared to CBC. more...

miRNA expression data from rhesus macaque postnatal brain in prefrontal cortex: time course

(Submitter supplied) Gene expression changes determine functional differentiation during development and are associated with functional decline during aging. While developmental changes are tightly regulated, regulation of aging changes is not well established. To assess the regulatory basis of age-related changes and investigate the mechanism of regulatory transition between development and aging, we measured mRNA and microRNA expression patterns in brains of humans and rhesus macaques over the entire species’ lifespan. more...

MicroRNA expression in primate postnatal brain through lifespan

(Submitter supplied) While multiple studies have reported the accelerated evolution of brain gene expression in the human lineage, the mechanisms underlying such change remain poorly understood. Here we address this issue from a developmental perspective, by analyzing mRNA and microRNA (miRNA) expression in two brain regions within macaques, chimpanzees and humans throughout their lifespan. We find that developmental profiles of trans-regulators, such as miRNA, as well as their target genes, show the fastest rates of human-specific evolutionary change. more...

MicroRNA expression and regulation in human, chimpanzee, and macaque brain

(Submitter supplied) Among other factors, changes in gene expression on the human evolutionary lineage have been suggested to play an important role in the establishment of human-specific phenotypes. However, the molecular mechanisms underlying these expression changes are largely unknown. Here, we have explored the role of microRNA (miRNA) in the regulation of gene expression divergence between adult humans, chimpanzees and rhesus macaques, in two brain regions: prefrontal cortex and cerebellum. more...

miRNA expression data from neonatal and adult human livers

Widespread splicing changes in human brain development and aging

(Submitter supplied) Differences in transcript splicing are well documented among tissues and between sexes in humans and other organisms. Further, splicing changes in selected genes have been reported in human and mouse brain development. Using high-throughput RNA sequencing we characterized the transcriptome-wide splicing changes occurring over the lifespan in the human brain. We found that in two brain regions, prefrontal cortex and cerebellum, as many as 37% of all expressed protein-coding multi-exon genes undergo significant splicing changes between birth and 98 years of age. more...

Deep sequencing of small RNAs from whole nematode C. elegans during aging

(Submitter supplied) MicroRNAs (miRNAs) have been implicated in gene regulation in many complex processes, including aging. Moreover, expression levels of many miRNAs are altered during aging, though we know little about the mechanisms that regulate this process. To address these issues, we investigated the network of regulatory factors that interact with miRNAs during aging. We found miR-71, a known promoter of normal lifespan, to be an especially important regulator of alg-1/Argonaute. more...

Alzheimer's Disease Dataset

(Submitter supplied) This dataset contains microarray data from normal controls (aged 20-99 yrs) and Alzheimer's disease cases, from 4 brain regions: hippocampus, entorhinal cortex, superior frontal cortex, post-central gyrus. Changes in expression of synaptic and immune related genes were analyzed, investigating age-related changes and AD-related changes, and region-specific patterns of change. These AD cases were processed simultaneously with the control cases (young and aged) included in GSE11882 (GSE11882 dataset contains data exclusively from normal control brains).

Gene expression changes in the course of normal brain aging are sexually dimorphic

(Submitter supplied) This dataset of cognitively normal controls is a subset of the GSE48350 dataset, which additionally contains microarray data from AD brains. Gene expression profiles were assessed in the hippocampus (HC), entorhinal cortex (EC), superior frontal gyrus (SG), and postcentral gyrus (PCG) across the lifespan of 63 cognitively intact individuals from 20-99 years old. New perspectives on the global gene changes that are associated with brain aging emerged, revealing two overarching concepts. more...

Gene expression data from temporal cortex of young adult, old and AD-like Microcebus murinus

(Submitter supplied) Aging is the primary risk factor of neurodegenerative disorders such as Alzheimer's disease (AD). However, the molecular events occurring during brain aging are extremely complex and still largely unknown. For a better understanding of these age-associated modifications, animal models as close as possible to humans are needed. We thus analyzed the transcriptome of the temporal cortex of the primate Microcebus murinus using human oligonucleotide microarrays (Affymetrix). more...
