Hepatocyte nuclear factor 4 alpha knockout effect on the embryonic liver

Analysis of embryonic day 18.5 livers lacking hepatocyte nuclear factor 4 alpha (HNF4alpha). Cre/lox system used to knock out Hnf4a in liver. HNF4alpha is a nuclear hormone transcription factor. Results provide insight into the role of HNF4alpha in hepatogenesis.

Comparison of HNF4 null mouse embryonic livers with control mouse embryonic livers

(Submitter supplied) To study the role of hepatic nuclear factor alpha (HNF4a in hepatogenesis, we used loxP-Cre technology to eliminate it from developing mouse livers. A comparison of control and experimental livers revealed that hepatocytes lacking HNF4a_failed to fully differentiate. Specifically, HNF4a null liver cells failed to express genes that are markers of mature hepatocytes including Gys2, Pck1, and G6pc. Gene expression in epithelial and non-epithelial cells of renal origin

(Submitter supplied) We aimed to define epithelial-specific genes in the kidney. In the developing mouse kidney at E12.5 epithelial cells are restricted to the ureteric bud, while mesenchymal cells surrounding the ureteric bud are non-epithelial. The mouse renal epithelial cell line mIMCD-3 was used to represent kidney epithelia in vitro. Gene expression was analyzed using Affymetrix microarrays in ureteric bud stalks, ureteric bud tips, and mIMCD-3 cells and compared to metanephric mesenchyme.
