Enhanced RNAi does not provide efficient innate antiviral immunity in mice in vivo. [RNA-Seq spleen]

(Submitter supplied) In RNA interference (RNAi), long double-stranded RNA (dsRNA) is cleaved by Dicer endonuclease into small RNA interfering RNAs (siRNAs), which guide degradation of complementary RNAs. While RNAi mediates antiviral innate immunity in plants and many invertebrates, vertebrates adopted sequence-independent response and their Dicer produces siRNAs inefficiently because it is adapted to process small hairpin microRNA precursors in the gene-regulating microRNA pathway. more...

Gene expression of Tamoxifen induced control (TC), dormancy-like (TD) or relapse (TR) cell samples

(Submitter supplied) To generate a dormancy state in vivo, female BALB/c athymic mice with ER+MC cell (MCF7) derived xenografts were treated with Tamoxifen (TAM). Throughout the treatment period, no significant differences in the body weight of the TAM-treated groups (TD and TR) compared to the vehicle-treated groups (TC) were observed. Whereas xenografts in the vehicle-treated group (as control group: TC) continued to increase in volume, the xenograft volumes of the TAM-treated group regressed 20%-30% in the first week and then remained stable during the four-week administration of TAM. more...

(Submitter supplied) To generate a dormancy state in vivo, ovariectomized female BALB/c athymic mice (simulating the post-menopausal state) with ER+MC cell (MCF7) derived xenografts were treated with Fulvestrant (FUL). Throughout the treatment period, no significant differences in the body weight of the FUL-treated groups (FD and FR) compared to the vehicle-treated groups (FC) were observed. Signal recovery in single cell batch integration

(Submitter supplied) Data integration to align cells across batches has become a cornerstone of single cell data analysis, critically affecting downstream results. Yet, how much biological signal is erased during integration? Currently, there are no guidelines for when the biological differences between samples are separable from batch effects, and thus, data integration involves a lot of guesswork: Cells across batches should be aligned to be “appropriately” mixed, while preserving “main cell type clusters”. more...

Obesity escalates Alzheimer’s disease and cancer via excess phosphatidylethanolamine-driven membrane remodeling

(Submitter supplied) The paradox of Alzheimer’s disease (AD) and cancer refers to the asynchronism of the two diseases. Understanding the correlation between cancer and AD could enhance research efficiency by consolidating efforts, lead to broader therapeutic impacts through drugs targeting shared mechanisms. Obesity is believed to synchronize this dichotomy by coordinating the onset and progression of both AD and cancer, although the precise mechanism remains unclear. more...

miR-193b-5p and miR-374b-5p are aberrantly expressed in endometriosis and suppress endometrial cell migration in vitro

(Submitter supplied) Endometriosis is a highly prevalent gynecological disease affecting 10% of women of reproductive age worldwide. miRNAs may play a role in endometriosis, though their exact function remains unclear. Here, we identified differentially expressed miRNAs in endometriosis and studied their functional role in the disease. (2) Methods: Endometrial tissue was collected from women with endometriosis (n=15) and non-endometriosis controls (n=17). more...

Elucidating human female genital tract resident DC response to HIV-1 exposure.

(Submitter supplied) The female genital tract (FGT) represents a complex and dynamic environment with specialized immune mechanisms uniquely designed to maintain a delicate balance between protection against invading pathogens and accommodating the unique physiological changes associated with reproductive function. Dendritic cells (DCs) are critical in shaping mucosal immunity against pathogens and maintaining tissue homeostasis. more...

Aging modifies endometrial dendritic cell function and unconventional double negative T cells in the human genital mucosa.

(Submitter supplied) Immune function in the genital mucosa balances reproduction with protection against pathogens. As women age, genital infections, and gynecological cancer risk increase, however, the mechanisms that regulate cell-mediated immune protection in the female genital tract and how they change with aging remain poorly understood. Unconventional double negative (DN) T cells (TCRαβ + CD4-CD8-) are thought to play important roles in reproduction in mice but have yet to be characterized in the human female genital tract. more...

The DNA demethylase TET1 regulates the impact of modified maternal folic acid status on embryonic brain development.

(Submitter supplied) Folic acid (FA) is well known to prevent neural tube defects (NTDs), but we do not know why many human NTD cases still remain refractory to FA supplementation. Here, we investigate how the DNA demethylase TET1 interacts with maternal FA status to regulate mouse embryonic brain development. We determined that cranial NTDs display higher penetrance in non-inbred than in inbred Tet1-/- embryos and are resistant to FA supplementation across strains. more...

Hypothermic oxygenated machine perfusion influences the immunogenicity of donor livers in humans

(Submitter supplied) Hypothermic oxygenated machine perfusion (HOPE) is an organ preservation strategy shown to reduce ischemia-reperfusion-injury (IRI)-related complications following liver transplantation this remains to be evaluated in humans. Analysis of liver tissue transcriptome at reperfusion revealed an effect of HOPE on the reactive oxygen species pathway. Two weeks post transplantation, HOPE recipients exhibited increased circulating CD4+FOXP3+CD127lo regulatory T cells (Tregs) (p<0.01), which corresponded to a higher frequency of donor specific Tregs (p<0.01) and was followed by reduced alloreactivity index of CD8+ T cells 3 months post-transplant. more...

ID1 and ID3 functions in the modulation of the tumor immune microenvironment in Adult Patients with B-Cell Acute Lymphoblastic Leukemia

(Submitter supplied) The diagnosis of B-Cell acute lymphoblastic leukemia (B-ALL) in adults often carries a poor prognosis. ID1 and ID3 genes have been identified as predictors of poor response in Colombian adult B-ALL patients, playing roles in cancer development. In different cancer models, these genes have been associated with immune regulator populations within the tumor immune microenvironment (TIME). B-ALL development alters the immune cell composition and the bone marrow (BM) tumor microenvironment, affecting disease progression and response to therapy. more...
