Maternal immune cell gene expression associates with maternal gut microbiome, milk composition and infant gut microbiome

(Submitter supplied) Background - Prepregnancy overweight and obesity promote deleterious health impacts on both mothers during pregnancy and the offspring. Significant changes in the maternal peripheral blood mononuclear cells (PBMCs) gene expression due to obesity are well-known. However, during pregnancy the impact of overweight on immune cell gene expression and its association with maternal and infant outcomes is not well explored. more...

Targeted gene expression panel for meningiomas

(Submitter supplied) A custom Nanostring nCounter panel of 100 or 75 meningioma related genes and reference genes was developed, and performed on human frozen and FFPE meningioma samples. This was done as part of a study developing and validating a targeted gene expression biomarker for meningioma outcomes after surgey and benefit from raditherapy.

Epigenetic aging of the demographically non-aging naked mole-rat [RRBS]

(Submitter supplied) The naked mole-rat (NMR) is an exceptionally long-lived rodent that does not show increased mortality with age, uniquely defining NMR as a demographically non-aging mammal. Here, we performed bisulfite sequencing on over one hundred NMR blood samples differing in age, assessing more than 3 million common CpG sites. We observed an information loss of the NMR methylome during aging suggesting that NMR ages. more...

Proteogenomic anlalysis of glioblastoma (GBM)

(Submitter supplied) To examine patterns associated with survival, we assembled a cohort of GBM patients whose survival ranges from less than 3 months and up to 10 years, most of which are not bearing isocitrate-dehyderogenase (IDH)-1 mutation and did not undergo prior treatment. We integrated high-resolution mass-spectrometry proteomics and RNA sequencing to examine the yet unresolved proteomic contribution to poor patient outcome, and compared it to the more established transcriptomic contribution and to published single-cell data. more...

Prevention of tuberculosis in rhesus macaques by a cytomegalovirus-based vaccine

(Submitter supplied) RhCMV-based TB vaccines are able to elicit and maintain immune effector responses that can control Mtb at the earliest stages of infection, and the protection afforded by this vaccine can be complete, if not sterilizing. To our knowledge, this is the first demonstration of complete prevention of TB disease in Rhesus Macaques (RM) by a long-acting vaccine after highly pathogenic (Erdman strain) Mtb challenge.

Expression data from human heart

(Submitter supplied) Global gene expression is altered in heart failure. This syndrome can be caused by cardiovascular diseases, including dilated cardiomyopathy (DCM), ischemic cardiomyopathy (ICM), hypertrophic cardiomyopathy, viral or toxic myocarditis, hypertension, and valvular diseases. We used microarrays to evaluate the impact of heart failure on human nucleocytoplasmic transport-related genes examining simultaneoulsly both dilated and ischemic human cardiomyopathies compared to normal hearts.

Dilated cardiomyopathy: left ventricle apex

Analysis of left ventricle (LV) from explanted hearts of male patients with dilated cardiomyopathy (DCM). DCM is characterized by systolic contractile dysfunction of the cardiac chambers. Results provide insight into molecular mechanisms underlying the changes in LV function observed in DCM.

tRNAs marked with CCACCA are targeted for degradation

(Submitter supplied) The CCA-adding enzyme adds CCA to the 3' ends of transfer RNAs (tRNAs), a critical step in tRNA biogenesis that generates the amino acid attachment site. We found that the CCA-adding enzyme plays a key role in tRNA quality control by selectively marking unstable tRNAs and tRNA-like small RNAs for degradation. Instead of adding CCA to the 3' ends of these transcripts, CCA-adding enzymes from all three kingdoms of life add CCACCA. more...
