Deletion mutant analysis of established glucose signaling and metabolic pathway members in Saccharomyces cerevisiae

(Submitter supplied) To understand the organisation of the glucose regulatory system, we analysed 91 deletion mutants of established glucose signalling and metabolic pathway members in Saccharomyces cerevisiae by DNA microarrays. These deletion mutants do not induce pathway-specific transcriptional responses reflecting the tight interconnection between pathways of the glucose regulatory system. Expression profiling of yeast kinase and phosphatase knockouts

(Submitter supplied) To understand relationships between phosphorylation-based signaling pathways, we analyzed 150 deletion mutants of protein kinases and phosphatases in S. cerevisiae using DNA microarrays. Downstream changes in gene expression were treated as a phenotypic readout. Double mutants with synthetic genetic interactions were included to investigate genetic buffering relationships such as redundancy. (Submitter supplied) 1. Oligos are resuspended and adjusted to 10µM in 150mM phosphatebuffer, pH8.5 in Genetix 384-wells plates 2. Array production is done using a Biorobotics MicroGridII spotter located in a low dust cleanroom (ISO7/ downflow ISO5) at 18C, at 48% humidity 3. Slide postprocessing is carried out according to CodeLink protocol: Printed slides are kept at 20C, 75% humidity for 24 hours 4.
