The gene expression profiling among normal chondrocyte, osteoarthritis chondrocyte and chondrosarcoma

(Submitter supplied) To investigate the gene expression profiling in bone diseases including osteoathritis (OA) and bone cancer (chondrosarcoma), different chondrocytes were isolated and assessed. For OA, the paired normal and OA chondrocytes were isolated from patient with total knee replacement surgery. Identification and delineation of dys-regulated genes between normal and OA chondrocytes and the regulatory mechanisms may provide the opportunity to treat OA. more...

Defective N-Glycosylation of IL6 Induces Metastasis and Tyrosine Kinase Inhibitor Resistance in Lung Cancer

(Submitter supplied) The biological consequences of various IL-6 glycoforms are unknown. To uncover the function of N-glycosylation of IL-6 in lung cancer, we compare the gene expression profiles in NSCLC cells induced by conditioned medium containing IL-6 with complete N-glycosylation (NG-IL6) or defective N-glycosylation (deNG-IL6).

Gene expression profiles of depletion of Daxx and HOTAIR

(Submitter supplied) In this study, we found that Daxx may modulate HOTAIR stability, and therefore, regulate H3K27me3 levels. To further examine the possible mechanism and biological consequence, we separately knocked down Daxx and HOTAIR with a lentivirus plasmid constructed with short hairpin RNA and siRNA oligonucleotides, respectively. We then used microarray analysis to identify several candidate genes regulated under the Daxx-HOTAIR axis.

HCT116 colorectal cancer cells treated with 2.5 mM Metformin

(Submitter supplied) Metformin is a drug used in the treatment of type 2 diabetes mellitus. Various studies have elucidated its anticancer properties. In this study, the effect of metformin on the differentiation and tumor microenvironment of colorectal cancer cells (CRC) was evaluated. For our study, we have used HCT116 colorectal cancer cell line and treated the cells with Metformin. Maximum tolerable non-toxic dose of metformin on HCT116 cells was determined by MTT assay. more...

Effects of Apigenin and Rutaecarpine on Gene Expression Profiling of Human BMSC-derived osteoblasts [Microarray]

(Submitter supplied) Aging causes dysfunctional changes to the bone microenvironment that lead to osteoporosis development. Osteoporosis is a serious bone health issue that is characterized by decreased bone formation and increased bone resorption. The aim of this study is to evaluate the effects of natural compounds, Apigenin and Rutaecarpine on osteoblast differentiation and examine if Apigenin and Rutaecarpine can rescue osteogenesis in aging-context. more...

Investigation of the anti-tumor mechanism of tirabrutinib, a highly selective Bruton’s tyrosine kinase inhibitor, by transcriptomics

(Submitter supplied) Tirabrutinib is a highly selective BTK inhibitor for treating hematological malignancies. This study analyzed the anti-tumor mechanism of tirabrutinib with microarray analysis of ABC-DLBCL xenograft model tumors. The comprehensive gene expression analysis revealed that tirabrutinib downregulated IRF-4, MYC, and mTORC1 pathway-related genes in TMD8, suggesting the crucial roles of these pathways in ABC-DLBCL.

Human MCF7 cells engineered to overexpress MYF6 vs. Control cells containing empty vector constructs [Rec2]

(Submitter supplied) Background: Proliferation is an important hallmark of cancer development and progression. Tumor growth rate significantly influences the course of the disease, and patients with tumors that have a high proliferation rate have an increased risk (10-20%) of metastasis and death. The genomic region encoding the transcription factor MYF6 is often altered in breast cancer, and alterations in MYF6 methylation have been linked to several different cancer types. more...

Analysis of Primary and Expanded NK Cells for Future Clinical Use

(Submitter supplied) Clinical Natural Killer (NK) cell therapy for the treatment of leukemia is currently limited by the availability of larger numbers of functional NK cells. In vitro NK cell expansion can be used to generate higher numbers of NK cells and shows the potential to improve the cytotoxicity against tumor cells. A critical need for the clinical application of manufactured NK cells is their molecular and functional characterization. more...

Inter-layer and inter-subject variability of circadian gene expression in human skin

(Submitter supplied) This work reports 24 h gene expression rhythms in two skin layers, epidermis and dermis, in a cohort of young, healthy adults, who maintained natural, regular sleep-wake schedules. 10% of the expressed genes showed such diurnal rhythms at the population level, of which only a third differed between the two layers. Amplitude and phases of diurnal gene expression varied more across subjects than layers, with amplitude being more variable than phases. more...

A clinical-grade HLA haplobank of human induced pluripotent stem cells derived from Japanese donors

(Submitter supplied) We constructed a clinical-grade haplobank of 27 induced pluripotent stem cells (iPSCs) lines prepared in accordance with good manufacturing practice regulations from seven donors who were homozygous for one of the four most frequent human leukocyte antigen (HLA)-haplotypes in Japan. The haplobank could provide HLA-matched iPSCs lines to ~40% of the Japanese population. Since the first release in 2015, these iPSC lines have been used in more than 12 clinical studies. more...

A Minimal Subset of Seven Genes Associated with Tumor Hepatocyte Differentiation Predicts a Poor Prognosis in Human Hepatocellular Carcinoma

(Submitter supplied) An integrative transcriptomics analysis was performed to evaluate the clinical relevance of genes associated with hepatocyte differentiation in human hepatocellular carcinoma (HCC). The well-established HepaRG cell line model was used to define a gene expression signature reflecting the status of tumor hepatocyte differentiation. This signature was able to stratify HCC patients into clinically relevant molecular subtypes. more...

Genes regulated by SMAD3 in HCC cells

(Submitter supplied) To identify genes regulated by SMAD3, we have employed whole transcriptome microarray expression profiling to identify genes downstream of SMAD3. Total RNAs from SMAD3-overexpression (7721-SMAD3 vs 7721-VEC) or SMAD3-knockdown (LM3-shSMAD3 vs LM3-CON) cell lines were measured.
