RNA seq data examining response to chemical treatment to build the first gene co-expression network of zebrafish

(Submitter supplied) This study combines RNA seq data from a number of different expeirments examining response to chemical treatment to build the first gene co-expression network of zebrafish. We interrogate this network to determine how the transcriptomic map responds to different types of chemicals and what pathways and processes are critical to chemical response

Coupling genome-wide transcriptomics and developmental toxicity profiles in zebrafish to characterize polycyclic aromatic hydrocarbon (PAH) hazard

(Submitter supplied) Polycyclic Aromatic Hydrocarbons (PAHs) are diverse environmental pollutants associated with adverse human health effects. Many studies focus on the carcinogenic effects of a limited number of PAHs and there is an increasing need to understand mechanisms of developmental toxicity of more varied yet environmentally relevant PAHs. A previous study characterized the developmental toxicity of 123 PAHs in zebrafish. more...

Alterations of DNA methlation in oral cavity of mice induced by dibenzo[def,p]chrysene

(Submitter supplied) Despite advances in treatment modalities, 5-year survival of OSCC has remained at ~50% for the past decades, mostly due to the high risk of developing secondary primary tumors. Early detection of OSCC represents one of the most promising approaches to improving survival. In this study, we examined the alterations of DNA methylation in early stage of oral carcinogenesis in mice induced by carcinogen dibenzo[def,p]chrysene. more...

Transcriptional Response of Male MutaTMMouse Spleen to Dibenzo[def,p]chrysene Exposure

(Submitter supplied) Dibenzo[def,p]chrysene (DBC) is the most carcinogenic polycyclic aromatic hydrocarbon (PAH) examined to date. We investigated the immunotoxicity of DBC, manifested as spleen atrophy following acute exposure of adult Muta™Mouse males by oral gavage. Mice were exposed to 0, 2.0, 6.2, or 20.0 mg DBC /kg-bw per day, for three days. Genotoxic endpoints (DBC-DNA adducts and lacZ mutant frequency in spleen and bone marrow, and red blood cell micronucleus frequency) and global gene expression changes were measured. more...

Polycyclic aromatic hydrocarbons as skin carcinogens: Comparison of benzo[a]pyrene, dibenzo[def,p]chrysene and three environmental mixtures in the FVB/N mouse

(Submitter supplied) The polycyclic aromatic hydrocarbon (PAH), benzo[a]pyrene (BaP), was compared to dibenzo[def,p]chrysene (DBC) and combinations of three environmental PAH mixtures (coal tar, diesel particulate and cigarette smoke condensate) using a two stage, FVB/N mouse skin tumor model. DBC (4 nmol) was most potent, reaching 100% tumor incidence with a shorter latency to tumor formation, less than 20 weeks of 12-O-tetradecanoylphorbol-13-acetate (TPA) promotion compared to all other treatments. more...

HepG2 treated with Polycyclic Aromatic Hydrocarbons (PAHs)

(Submitter supplied) Assessing the potential carcinogenicity of human toxins represents an ongoing challenge. Chronic rodent bioassays predict human cancer risk with limited reliability, and are expensive and time-consuming. To identify alternative prediction methods, we evaluated a transcriptomics-based human in vitro model to classify carcinogens by their modes of action. The aim of this study was to determine the transcriptomic response and identify specific molecular signatures of polycyclic aromatic hydrocarbons (PAHs), which can be used as predictors of carcinogenicity of environmental toxins in human in vitro systems. more...

FTC-238/hrTPO/RSK008 treated with thyroid peroxidase-disrupting chemicals

(Submitter supplied) These experiments have systematically assessed the potential utility of transcriptomic endpoints as enhancements to the guaiacol assay. Previously, we demonstrated that benzophenone-2, benzophenone, perfluorooctane sulfonate, bisphenol A bis ether, and vinclozolin decreased TPO activity, and that dibutyl phthalate, carbaryl, dibenzo(a,h)anthracene, benzo(a)pyrene, and methylmercury increased TPO activity. more...
