Study: Transcriptome profiles of antibody-producing CHO cells across different amplification strategies
show Abstracthide Abstract
The mechanism of action behind the increased secretion capacity of the amplified progeny of CHO cells is investigated in this study through comparative transcriptomic analysis Overall design: The amplified DG-44 CHO cells, taken at both exponential and stationary growth phase were compared to non-amplified CHO DG-44 cells. Non-producing CHO-S samples were used as negative controls.
Sample: CHO cells, A03_EXP_Duplicate

Library:
Instrument: Illumina MiSeq
Strategy: RNA-Seq
Source: TRANSCRIPTOMIC
Selection: cDNA
Layout: SINGLE
Construction protocol: Cell cultures were re-suspended in RLT buffer (Qiagen) and kept at −80 °C until RNA was extracted using the RNeasy kit (Qiagen) and on-column DNAse digestion. Library preparation was performed with Illumina's TruSeq Stranded mRNA Library Prep Kit High Throughput (Catalog ID: RS-122-2103), according to manufacturer's protocol
