SRX23390116: Whole genome sequencing of V. fischeri strain derived from ES114

Design: Genomic DNA from V. fischeri strains was extracted using the Invitrogen PureLink Genomic DNA Mini Kit. DNA was sonicated to generate fragments of approximately 500 bp, and gDNA libraries were prepared using the NEBNext Ultra II DNA library prep kit for Illumina, which includes end-repair, adaptor ligation, and addition of index primers. Sequencing was performed by the University of Georgia Genomics and Bioinformatics Coreon an Illumina NextSeq500 instrument.

Study: Vibrio fischeri D-glutamate suppressor generation and characterization

This study used D-glutamate auxotrophic mutants of Vibrio fischeri to study the evolution of peptidoglycan biosynthesis and structure. Suppressor mutants were generated by growing an auxotrophic strain without necessary exogenous D-glutamate. Mutants were then characterized and genome sequenced, in order to determine the mechanism of suppression.
