show Abstracthide AbstractIn order to investigate the molecular basis of carotid body chemoreceptor sensitisation in the Spontaneously Hypertensive Rat (SHR) we have sequenced the transcriptomes of bilateral carotid body samples from aged-matched, male SHR and control Wistar-Kyoto rats. Overall design: Bilateral (right and left) carotid body samples of n=6 individual SHR/NHsd and WKY/NHsd (male, 12-14 weeks old) rats were microdissected from common carotid artery bifurcations following intracardial perfusion with ice-cold PBS. Total RNA was isolated using phenol-chloroform phase seperation method and Zymo Directzol RNA Microprep kit. Average RNA Integrity Number (RIN) of samples processed for sequencing (n=24) was 7.67. Poly-A enriched sequencing libraries were constructed using TruSeq® Stranded mRNA library preparation kit (20020594, Illumina). Libraries were pooled and sequenced on Illumina NextSeq500 platform using NextSeq 500/550 High Output v2.5 (150 Cycles) Kit (20024907; Illumina) on a paired-end multiplex mode.
