The 4 Ago antibodies were obtained from our cooperation partner Prof. Dr. Gunter Meister from University Regensburg and were manufactured in house. TRIzol extraction of Ago- / isotype control-associated and total RNA was performed according to the manufacturer's instructions.

Biotinylated aRNA was prepared according to the standard Affymetrix protocol from (GeneChip 3' IVT Express Kit Manual, 2008-2009, Affymetrix).

Hybridization protocol |
Following fragmentation, aRNA was hybridized on Affymetrix Human Genome U133 2.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450 (performed by the core facility BMFZ Düsseldorf).

GeneChips were scanned using the GCS 3000 from Affymetrix (performed by the core facility BMFZ, Düsseldorf).
