Cryopreserved tissue was thawed in a 37°C water bath for 75 seconds. Nuclei were released from the tissue via dounce homogenization using a buffer containing Triton X-100 (0.1 v/v). The homogenate was filtered through a 10µM cell strainer and nuclei were pelleted (500 x g for 5 minutes at 4°C) and washed before being resuspended in 1% w/v BSA, 1X PBS, and 0.2 U/µL RNase inhibitor
Library was constructed according to manufacturers instructions (single cell 3' v3.1, 10x Genomics).
