Mice of at least 24 weeks old where firstly castrated, and 72h later, they received sub-cutaneous injections of either vehicule (1:10 ethanol 96% / vegetable oil), Estradiol (30 µg/g of mouse weight) and/or Testosterone (30 µg/g of mouse weight). Estradiol will activate the estrogen receptor alpha (ERa), whereas Testosterone will activate the androgen receptor (AR).

Thirty mice in total (16 C57BL/6J Ptenfl/fl [normal or WT] and 14 C57BL/6J PB-Cre4+/-;Ptenfl/fl [prostate cancer]) were bred and housed in a 12 hour light, 12h dark cycle at 22°C.

24 hours after injections, mice prostates were harvested, and RNA was extracted and purified with the QIAGEN RNeasy Mini Kit.
mRNA library was generated using NEBNext Ultra II Directional RNA library prep kit.
