Treatment protocol TPC-1 cell line cells were plated at 1x10^5 per 12-well dish and after 24 h transfected in quadruplicate with PTCSC3 expression vector and with empty vector (pcDNA3) as control using Lipofectamine2000 reagent (Invitrogen).
Growth protocol TPC-1 cells were cultured in DMEM (Gibco) supplemented with 10% fetal calf serum (Gibco) and 1x MEM non-essential amino acids (Gibco) at 37C in 5% CO2.
Extracted molecule total RNA
Extraction protocol At 24 h after transfection the cells were washed once in PBS and total RNA was extracted (Trizol reagent, Invitrogen) and DNase-1 treated (Ambion).
Label Cy3
Label protocol RNA samples were labeled with cyanine-3 (Cy3) using the Agilent Low Input One–Color Labeling kit (Agilent Technologies, CA).
 
Hybridization protocol Samples were hybridized to the array for 17 hours at 65°C according to the manufacturer's protocol.
Scan protocol Slides were scanned with an Agilent G2505C Microarray Scanner.

Data processing Signal intensities were detected by Agilent DNA Microarray scanner and quantile normalized with R software using 'limma' bioconductor library.
