Extraction protocol |
The transfected MDA-MB-231 cells were grown in phenol-red free RPMI with 5% CDFBS before subjecting to 10nM E2 or vehicle treatment. Total RNA was harvested at day 2 and 10 using Trizol reagent (Invitrogen) followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.

The expression intensities after E2 treatment were normalized with the expression intensites before E2 treatment at the same probes from the same replicate. The log ratios of normalized expression intensities were averaged from different repicates. The avareged log ratio of the normalized expression intensities were averages from the multiple probes to generate the final expression level for the same gene. [Average log ratio data of the expression intensities from the same gene before and after E2 treatment is provided as a Series supplementary file]
